Infectious diseases are common diseases in clinic.Infection often leads to an increase in the number of white blood cells,as well as an increase in the concentration of C-reactive protein(CRP)and Serum Amyloid A(SAA).At present,white blood cell testing,CRP testing items and SAA testing items are generally tested separately by different instruments,so combining these three testing items together can improve the testing efficiency and facilitate the clinical diagnosis of infectious diseases.So,this paper completed the development of a hardware system that combines the functions of the three into the same instrument.The main work of this paper includes the following three aspects:1.Combined with the gas circuit,liquid circuit and mechanical structure involved in the pretreatment process and the measurement process of blood samples,a hardware system based on MCU chip and FPGA chip is designed.At the same time,according to the different pretreatment and measurement methods of blood samples,the hardware system is modularized according to function,and the top-level design of the hardware system is completed.2.In this paper,a control board is designed to coordinate other circuit boards and modules,which is the core and top layer of the hardware system.The control main board is responsible for executing commands and uploading measurement issued by the host computer software system,and for the orderly operation of other modules in the control hardware system.The control board makes full use of the rich gate logic unit,embedded RAM(random dynamic access memory),clock resources,etc.in the FPGA chip.The FPGA collects caches and transmits data from the laser scattering signal detection module and the Coulter signal detection module.In addition,according to the combined detection method of laser scattering and cytochemical staining for the classification of white blood cells,a laser scattering signal detection module is designed.The detection module collects the scattered light from two angles of white blood cells,and then the white blood cell will be classified by classification algorithm according to the scatteredlight intensity of these two angles.At the same time,the Coulter signal detection module is designed according to the Coulter detection principle of white blood cell count,red blood cell and platelet count for blood cell count.In addition,the protein measurement module is designed according to the immunoscatter turbidimetry method for protein measurement,which is used to detect the concentration of CRP protein and SAA protein.3.In this paper,the data transmission performance of high-speed data acquisition module is tested and verified by validation data which is transmitted from the high-speed data acquisition port to the MCU chip memory,and the result transmission speed can reach 45MB/s,and the data is accurate.The maximum value of the stray light of the laser scattering signal detection module does not exceed 19,the fluctuation range is small,and its CV value does not exceed 0.6%.The laser scattering signal module is tested with 7μm counting microspheres,and the CV values ??of the high and low angle scattering signal peaks are 6.82% and 8.30%.The maximum noise of the Kurt signal detection module does not exceed 10,and the average value does not exceed 5.The Kurt signal detection module is tested with 7μm counting microspheres,and the average value of the Kurt signal peak is 183,and the CV value is 10.23%.The ratio of the stray light of the protein measurement module to the AD value of the reagent detection is about1.58%.The module by using 3 concentrations of 3 mg/L,10 mg/L,and 100 mg/L calibrators.The CV of the results of 10 tests Less than 10%.This result meets the CV requirement for the C-reactive protein assay kit proposed by the China Food and Drug Administration with a CV of no more than 10% repeatability.In the concentration range of 3~156mg/L,the measurement results have a good fit,and its linear correlation coefficient |r|=0.99243.This results meet the industry standard of C-reactive protein test kit proposed by the State Food and Drug Administration,and the CV is not more than10% repeatablility.
