Objective Compare the performance of botany properties,biological character,economic traits and quality characteristics according to the data of trial and select the one which gets high yield,steady character,and strong resistance for disease and is suitable for cultivation in our province.And to get the best part of the plant for DNA extraction,the best dosage of β ME and RNaseA and optimize the DNA extraction technique,different parts of plant,different dosages of β ME and RNaseA was tested.Methods 1.Random blocks design were taken to study on the characteristics of Amorphorphallus konjac.Each strain was a treatment and repeated 3 times.The field management unified.Different investigation method was taken on the basis of observation items.2.Based on the different dose of anti-oxygen and RNaseA and different parts of the plant,the orthogonal design was performed to optimize the condition for DNA extraction.Results 1.The study of character results showed as followed:These four kinds of konjac growth period were relatively close,the shortest growth period was Hubei strain;as plant traits,Hubei strain was the highest with its petiole thickest,the amount of leaves most and type of plant T,while the other strains had each other’s advantages,for example,Jiangzhong strain was the best from the comparing in the performance of tuber character.In the field of biological character,the Hubei stain was the highest and the Yu Nan strain was the lowest from the comparing in the rate of germination and Yu Nan strain was the highest from the comparing in the incidence of soft root disease.In term of economic traits,Hubei strain won the first place and the Yu Nan strain occupied the last place from the comparing of product;Jiangzhong stain was the largest and Yu Nan strain was the smallest from the comparing of growth coefficient.In the light of quality characteristics,Yu Nan strain was the best,Hubei strain occupied the second place,Jiangzhong strain got the third place,Xi Chang strain was in the last place from the comparing of percentage of KGM.2.The study of DNA extraction technology results showed as followed:The root was the best part of the plant for DNA extraction,the best dosage of β ME and RNaseA was 2%and 5 μL.Conclusion 1.In consideration of characters all above,we screened out Hubei strain which got high yield,steady character,and strong resistance for disease and was suitable for cultivation in our province.2.By testing the different parts of the plant and different dosage of β ME and RNaseA,we got the best condition of DNA extraction and we have cleared the obstacle for further study.
