After producing Konjac oligosaccharide (KOS) with fermentation enzyme-hydrolyzed Konjac glucomannan (KGM), this study identified the antioxidant ability of KOS through in vitro and in vivo. In vitro we studied the scavenging capacity of free radicals and its protection against DNA and protein damage. And then we examined the level of MDA and activity of SOD and GSH-PX in liver and plasma of Balb/c mice after feeding 21 days. On the base of its antioxidant ability, we synthesize Se-KOS with KOS and selenium, try to find out the chemosynthesis discipline, optimize synthesis condition. Take the trial production result as basis, optimize the enlarging synthesis technological conditions, and probe out a set of Se-KOS chemosynthesis project. Besides, Se-KOS has been analyzed with structure and common property. Try to provide theory basis and technical instructor for the production of Se-KOS in industry.With Konjac oligosaccharide (KOS) by enzyme-hydrolyzed Konjac glucomannan (KGM) for materials, we study the scavenging capacity of free radicals with a series projects and its protection against DNA oxidation damage with Agarose gel electrophoresis. Its protection against protein oxidation damage has been studied with quantity of carbonyl ramification which is produced by amino acid oxidation by active oxygen.Base on the antioxidant ability of KOS through in vitro scavenging free radicals and protection DNA and protein damage. Through feeding mice for 21 days to study the effects of KOS on plasma and liver antioxidant enzymes in miceAfter identification the antioxidant ability through in vitro and in vivo, we synthesize Se-KOS with KOS and selenium and optimize chemosynthesis condition. Then conclude the modality of Selenium with ultraviolet and infrared scan.Conclusions show that Konjac oligosaccharide has free radicals scavenging capacity and can protect DNA and protein damage caused by hydroxyl radicals; it reduced the level of MDA in liver and increased the activities of SOD and GSH-Px in liver and plasma of mince; KOS and selenium can be combined organically, the optimum condition is as below: KOS 1.00 g, Selenium Reagents 0.50 g, low speed stir, time 14 h , temperature 40℃, then dialysis time 30 h, low temperature lyophilizations; Inorganic selenium can be wiped off with chemicals in large-capacity production of Se-KOS, which avoided the bothering of dialysis bag and saved cost. So it makes the composing reaction being easy for industrialization production. Through with ultraviolet and infrared scanning, Se-KOS is organic selenium compounds of high se-containing.
