| Atractylodes lancea(Thunb.)DC.is perennial herb in Compositae and has effects of dispelling wind and cold,dampness and spleen,and eyesight and so on.It’s a common-used clinical medicine crude drugs.A.lancea is mainly produced in Jiangsu,Hubei,Anhui and Henan Province.Maoshan area of Jiangsu Province has long been recognized as genuine producing area.There are many problems in cultivation process,such as the confusion of varieties and different varieties of quality,the unsteadiness of the seedling quality and drug quality,the extensive management and low intensive degree,the unstable of yield and weak stress-resist ability,and continuous cropping obstacle,etc,which seriously restrict the quality and yield of A.lancea.Therefore,this paper studied on the key technology of good agriculture practice of A.lancea by used biotechnology,agricultural technology,modern analytical technique.This paper hoped to provide scientific basis for high quality,high yield and stable yield of A.lancea,and provide high-yielding herd for clinical.The research mainly included the following contents:(1)Comprehensive evaluation of stress resistance and active ingredient concentration of A.lancea from different habitat According to the characteristic that A.lancea likes coolness and dryness,and hates sultriness and dampness,the physiological indexes,photosynthesis parameters and chlorophyll fluorescence parameters of A.lancea from different habitats were studied under high temperature and waterlogging stress.Besides,this paper also determined the active ingredient concentration of A.lancea.The results showed that A.lancea from Jurong city showed the strongest ability to resistant to adversity stress.And medicinal material from Jurong city obtained best result too.(2)Researches on yield and the active ingredient concentration of A.lancea under different intercropping pattern According to the characteristic that A.lancea hate sultriness and sunlight-exposure,this paper designed that A.lancea intercroped with shrub(like Euonymus alatus and Gardenia)which had different branch spread and existed in base,and design monoculture as the control.This paper statisticsed fresh weight,dry weight and conomic benefit.Moreover,the marker component,cluster analysis and quality evaluation of A.lancea was determined by HPLC method.The results showed that gardenia-intercropping obtained highest yield,the highest atractylodin content and comprehensive evaluation result.intercropping mode had little effect on the active ingredient of A.lancea.But gardenia-intercropping showed the best conomic benefit.(3)Researches on yield and the active ingredient concentration of A.lancea under different key cultivation techniques According to several common way of field management,"3414" fertilization program,weeding and picking.Bud test were studied on A.lancea,and statistical methods was same as(2).The results showed that "N2P2K2"treatment group,"April weeding,July no weeding and September weeding" treatment group and "August excising all flower bud" treatment group obtained highest yield.And the effect of above field management on the active ingredient of Atractylodis Rhizoma were consistent with the yield results.(4)Researches on yield and the active ingredient concentration of A.lancea under different harvesting time and initial processing According to the fact that harvesting time of A.lancea is amphibolous in pharmacopoeia,and initial processing experience is inconsistency in different habitats,the harvesting time of A.lancea was studied from the October of second year to February of third year,and the drying temperature of A.lancea from same harvesting time was studied too.Moreover,the marker component,cluster analysis and quality evaluation of A.lancea was determined by HPLC method.The results showed that the A.lancea digged in December of second year showed most active ingredient,and highest yield,and 35℃ drying temperature was more appropriate.(5)Standard operating procedure(SOP)of standard cultivation on A.lancea After more than two years of special research on planting technique about A.lancea,this paper carried out to grope for high yielding and high qualitying planting technique.In order to provide scientific basis for practical production,this paper studied and worked out standard operating procedure(SOP)of standard cultivation on A.lancea,based on the previous studies.(6)Study on tissue culture and rapid propagation of A.lancea Based on the previous studies and present situation of industrial seedling,extensive studies had been conduced on disinfection method,induced proliferation,root induction of explants from A.lancea.And improvement program had been optimized.In order to provide scientific basis for achieve the industrial seedling and finally realize a way to provide rapid growth,high quality,low-cost A.lancea seedlings for bases,extensive studies had been conduced on training plantlet. |
