In this experiment,Tetrastigma hemsleyanum were used to study the changes of growth index under red,blue and white light treatments,such as stem length,diameter,fresh stem diameter,leaf area,microstructure of stem and leaf.As well as chlorophyll content,soluble protein,soluble sugar,antioxidant enzyme activity,MDA content,endogenous hormones and other physiological characteristics.And to study the inhibitory effect of Tetrastigma hemsleyanum ethyl acetate extract on HepG2 and SMMC-7721 liver cancer cells in vitro.The results show that:1.Under different light quality,the growth of stems and leaves of Tetrastigma hemsleyanum was inconsistent.Red light could stimulate the elongation of stem and the length of new stem was significantly higher than that of the control.There was no obvious promotion on stem diameter,leaf area and fresh weight.Under the blue light treatment,the diameter of new stems,leaf area and fresh weight of leaves were significantly higher than that of the control.Blue light could promote the expansion of the internal structure of new stems,increase the thickness of midrib and assimilation tissues,and promote the growth of Tetrastigma hemsleyanum.Three kinds of light quality on the diameter were no significant difference.2.Chla content and chla+b were the lowest under the red light treatment.Blue light was beneficial to increase the content of chla,the chlb content and chla+b of the control group were higher.The content of carotenoid in red light at the later stage of experiment was significantly higher than that in the control group.The chla/b under blue light was significantly higher than that of the control,showing the sun plants.The red light treatment of the chla/b value was low and show the characteristics of shade plants.Blu-ray increased the content of soluble protein and soluble sugar of Tetrastigma hemsleyanum,and the red light had less effect on promoting the accumulation of carbohydrate than blue light.The activity of SOD and POD under blue light was higher than that in control,the content of MDA was higher and the red light was lower.The effect of red,blue and white light qualities on CAT activity was insignificant.3.Red light was beneficial to the accumulation of IAA content and GA content in stems and leaves of Tetrastigma hemsleyanum.IAA and GA contents in stems and leaves were lower under blue light,but CTK and ABA contents were higher under blue light.There was a significant positive correlation between the length of stems and the content of GA,and the new stem diameter,leaf area and leaf thickness were positively correlated with CTK content.4.Tetrastigma hemsleyanum extract of ethyl acetate has inhibitory effect on proliferation and induction of apoptosis on liver cancer cells,but the cell cycle changes are not obvious;Tetrastigma hemsleyanum ethyl acetate extract can induce apoptosis by regulating the protein expression,the effect of inhibition on tumor cell proliferation was remarkable.In summary,the red light can promote the elongation of stems,blue light is conducive to plant sturdy and benefit to the accumulation of chlb,soluble protein content and soluble sugar content,and improve plant antioxidant capacity.The red light promoted the accumulation of IAA and GA,and the contents of CTK and ABA were higher under blue light.The ethyl acetate extract of Tetrastigma hemsleyanum had inhibitory effect on the proliferation of tumor cells.
