| In order to research the mechanism of Paulownia seed’s germination and differential expression of phytochrome gene(PHYA,PHYB)with different light qualities and temperatures,in the studies,the Paulownia seed was used as the research object and treated under different light(Red Light,Far-red Light,Blue Light,Purple Llight,White Light)and temperatures(5℃、15℃、20℃、25℃),to analysis it’s germination rate statistics,the Real-time PCR analysis was applyied to analyze Paulownia seed’s quantitative expression of phytochrome gene(PHYA,PHYB).The main findings were as follows:1.The influence of temperature and light quality on the germination of Paulownia seeds.(1)Paulownia seed has the highest germination rate at 25℃,which was a strict light demanding seed.Under dark conditions,seeds did not germinate without cryogenic treatment.After cryogenic treatment,there were signs of germination,which was most obvious after pre-chilling for40 days,but the germination rate is still not high,is 9.67%.(2)Without cryogenic treatment,It was not sensitive to short time light treatment at 15 and 20℃,not germinating.Under the condition of 25℃,the effect of red light irradiation was obvious in the short time,seed germination was achieved when irradiated with 1H.When irradiated with 12 h,it reached 57.67%.Germination rate increased with the increase of illumination time,and the effect was better than that of white light treatment.(3)With cryogenic treatment,the seeds of different light quality and light treatment significantly improved sensitivity.Light 1H,different light quality had seed germination and seed germination without cold treatment were significantly different.In addition to the FR light,the germination rate of blue light and purple light was significantly increased.The maximum germination rate reached at 12 h after pre-chilling for 20 days and 12 h after pre-chilling for 40 days.Red light treatment was most obvious to promote seed germination,The germination rate of 1h light treatment after pre-chilling for 20 days,was 47%,which was higher than white light irradiation 1h.Under the condition of continuous light irradiation,the germination rate reached the highest 90.33%,which was higher than that of white light.2.The influence of alternate treatment of R light and FR light on Seed Germination.(1)The effect of R light and FR light on the germination of Paulownia seeds wasreversible.FR light can reverse the effect of R light on the germination of Paulownia seeds,and R light can counteract the inhibitory effect of FR light on the germination of Paulownia seeds.The ability of FR light to reverse R light to promote seed germination decreased with the increase of the number of turns.(2)No matter whether the last illumination treatment was red or far red,The germination rate of Paulownia seeds increased with the increase of intercrossing times.The number of treatments to reach the highest germination rate reduced with increasing the illurmination time.3.The temperature and light quality on Paulownia seed phytochrome gene(PHYA,PHYB)differential expression analysis.(1)Without cryogenic treatment of Paulownia seeds in different light irradiation treatment,The expression levels of PHYA and PHYB were higher than those without light treatment.The most obvious change is white light.(2)With cryogenic treatment of Paulownia seeds under the dark conditions,The expression of PHYA and PHYB increased significantly compared with those without dark treatment.The difference was significant in 20 days.(3)Paulownia seeds treated by low temperature,Different light irradiation treatment compared with no light treatment,the expression of PHYB and PHYA in the light of 1H was greatly lowered.After pre-chilling for 20 days and after pre-chilling for 60 days showed that the amount of PHYA and PHYB expression was significantly different in seeds irradiated with no light when irradiated by 1h. |
