Study Of Gh Transgenic Channel Catfish,Ictalurus Punctatus,P1 And F1 Generation And Comparisons Of GH Transgenic Channel Catfish And GnRH Gene Knockout Channel Catfish | | Posted on:2020-04-01 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y L Zhang | Full Text:PDF | | GTID:2493305900985449 | Subject:Clinical Veterinary Medicine | | Abstract/Summary: | PDF Full Text Request | | Channel catfish is one of the most common farmed fish in north America.Transgenic technology has brought much convenience to people’s life since it was developed in 1980 s.The synthesis and secretion of growth hormone are closely related to the development and growth of fish.The overexpressing of growth hormone gene can promote fish growth.There is still some concern that transgenic fish may have a negative impact on ecosystem once there are some escapees.In this experiment,the P1 generation of transgenic channel catfish was generated through electroporation.F1 generated was generated by mating P1 transgenic channel catfish with P1 and P1 generation with blue catfish to combine the superiority of hybrid catfish and transgenes.The positive rate of P1 generation through electroporation can reach 15.15% but with apparent mosaicism.At the age of 6 months old,P1 AFP-cc GH channel catfish showed growth enhancement.At 12 months old,P1 transgenic channel catfish showed growth enhancement.At 6 months old,F1 generation of transgenic channel catfish showed growth enhancement.In addition,at 12 months old,there is no significant difference between the growth rate of transgenic channel catfish and that of controls.At the same time,the growth rate of 6-month-old hybrid catfish are much higher than the controls.At 12 months old,there is no significant difference between the growth rate of transgenic channel catfish and that of controls.At the same time,the growth rate of transgenic hybrid catfish was much higher than non-transgenic channel catfish controls.Growth hormone transgenic channel catfish,transgenic sterilization channel catfish,and non-transgenic channel catfish were cohabitated at high density,similar to a modular catfish culture system.Fish were harvested at 4 years of age and at 5 years of age after being communally grown in the same pond.Sex-age interactions were observed.CAB(tilapia Gn RH antisense transgene)females had the highest observed early body weight but were the smallest genetic type of female at final harvest.PAB(rainbow trout Gn RH antisense transgenic)had the highest observed body weight at final harvest for females.GAD(goldfish glutamate decarboxylase transgene)had the highest early body weight for males.AFP(channel catfish growth hormone gene transgenic)had the highest observed final body weight and the highest observed body weight gain between years 4and 5.Sexual maturity was highly and positively correlated with growth rate.Gn RH antisense transgenic channel catfish had reduced growth rate and sexual development.Alternative genetic sterilization techniques or the current technology coupled with selection may be needed to overcome this negative pleiotropic effect.The main points of this research project was that the superiority of selection,transgenic research and interspecific hybridization were combined with the weakness of each technology being prevented. | | Keywords/Search Tags: | Channel Catfish, Growth Hormone Gene, Transgenic Sterilization, Sex Maturity Score, Growth Rate | PDF Full Text Request | Related items |
