| Chinese prickly ash(Zanthoxylum bungeanum Maxim)is one of the ancient cultural economic plants originating from China.Z.bungeanum has a high economic value of flavoring,anticorrosion,health care,appetite increasing,anticancer,anesthesia and analgesia.The multiple function of Z.bungeanum indicated the importance of Chinese prickly ash industrial development.Sichuan is one of the main areas of Chinese prickly ash planting,in order to have a detailed understanding of the status quo of Chinese prickly ash diseases,we conducted a field survey on the area of Hanyuan County Yanyuan County,Renshou County,Pingchang County,Santai County of Sichuan Province,and identified pathogens and analyzed the pest risk of newly discovered virus.According to the investigation,there are 9 main diseases of Z.bungeanum which are mainly leaf disease and trunk disease.The highest frequency of Z.bungeanum is rust disease,which is the main disease of leaf.Plaster disease is the main disease of trunk and root rot is the only one disease found in root.A new type of prickly ash phytoplasma disease was found through investigation,which leaded the staminate flower blossoming,thus,the whole plant beared no fruit.The plant disease of Z.bungeanum is a new virus found in the process of this investigation identified as a pathogen.The universal primers of phytoplasma 16 Sr RNA were used to detect the susceptible plants of Z.bungeanum by nested PCR.The specific bands of1.2 KB was amplified and the sequences were homologous with the phytoplasma and showed highest homology with the 16 Sr V group of phytoplasma.It is indicated that the pathogen of Z.bungeanum has phytoplasma.It also indicates that nested PCR is suitable for the detection of phytoplasma diseases of Z.bungeanum,which provided technical guidance and theoretical proof for future detection.For the new found phytoplasma disease,using qualitative analysis and quantitative analysis,calculating the risk of harmful biological phytoplasma disease value of R=2.72 with multi index comprehensive evaluation method,we determinate that the phytoplasma disease is a highly dangerous forest pest. |
