Poria Cocos(Schw.)wolf is the dry sclerotia of Poria Cocos(Schw.)wolf,which was first recorded in Shennong herbal classic.It is listed as the top grade.It has the functions of invigorating the spleen,promoting water and moistening,and calming the heart.It is mainly used for the following diseases: less edema and urine,dizziness and palpitation of phlegm,less deficiency of spleen and food,loose stool and diarrhea,uneasiness and insomnia.Modern studies have shown that Poria cocos has diuretic,tranquilizing,bacteriostatic,enhancing the body’s resistance to disease and reducing blood sugar and other pharmacological effects.Poria cocos,as a large variety of traditional Chinese medicine,is widely used in clinical medicine,traditional Chinese medicine preparation,health food and cosmetics.The artificial cultivation of Poria cocos has a history of more than 1500 years,with a wide range of production areas,mainly distributed in Hubei,Hunan,Anhui,Yunnan and other places.The major production areas are still dominated by block wood cultivation,and the bag material cultivation aimed at reducing the use of wood is in the research stage.The planting area of Poria cocos covers a wide range,with different planting species,methods,climate and quality.At present,the quality standard of Poria cocos in the first volume of Chinese Pharmacopoeia(2015 Edition)stipulates the detection methods of alcohol soluble extract and ash,as well as some folk experience classification methods,which lack the data support of modern scientific instrument analysis.In this paper,the following studies have been carried out on the cultivation mode and quality control of Poria cocos as follows:(1)on the basis of the previous experimental research,the comparative study on the cultivation mode of Poria cocos as a substitute material is carried out,which is divided into two groups,i.e.the group with and without section wood in bag material,and the differences between the two cultivation modes are analyzed from the aspects of yield,survival rate and quality.The results show that each quality In the test data,the group of adding segment wood in bag material is better than that of not adding segment wood,which lays a foundation for the promotion and application of the later generation material cultivation.(2)HPLC-ELSD method for the determination of monosaccharide in Poria cocos water soluble polysaccharide was established.The precision,stability,repeatability and recovery of the method are all in accordance with the provisions of Chinese Pharmacopoeia.The advantages of this method are that the sample can be determined without derivatization,and the monosaccharide composition in the polysaccharide can be analyzed qualitatively and quantitatively quickly,and the method has strong universality and good repeatability,The results showed that there was no significant difference in the monosaccharide content of Poria cocos.(3)A method was established for the simultaneous determination of 7 triterpenoids in Poria cocos,i.e.dehydrotomorphic acid,polyporhinic acid C,3-epidehydrotomorphic acid,3-o-acetyl-16 α-hydroxy-hydrocortisonic acid,dehydrotomorphic acid,Poria cocoic acid and new acid of Poria cocos.The method has good precision,stability,repeatability,recovery rate,durability of correction factor,etc.it can be used for the simultaneous determination of dehydromotic acid,polypyrrhizic acid C,3-epidehydromotic acid,3-o-acetyl-16 α-hydroxy-hydrogenated Poria acid,dehydromotic acid,Poria acid,and new Poria acid in Poria cocos,and for the determination of several samples from different places The total content of triterpenoids in Poria cocos was not significantly different.(4)Based on the theory of traditional Chinese medicine,the model of "Spleen Deficiency" in rats was established to verify the effectiveness of water-soluble polysaccharides and triterpenes,the main quality control components of Poria cocos.The results showed that Poria cocos water-soluble polysaccharides and triterpenes had different degrees of improvement on the weight,swimming time,gastrointestinal absorption function,and various indexes in serum of rats with spleen deficiency,indicating that Poria cocos quality control components It has pharmacological activity and provides a new basis for the improvement of Poria cocos quality system.
