In this experiment,the natural wild semi-woody branches of Sabina vulgaris were used as materials,aged from two to three years branch.The experiment explored and screened the cutting propagation technology,from the selection of cuttings,the treatment of cuttings,the cuttage matrix and so on.The external morphology of cuttings was also observed in the experiment,and the internal mechanism of Sabina vulgaris rooting in cuttage was preliminarily explored,and the contents of cuttings under different substrates were analyzed.The purpose of this study was to find out the physiological basis of cutting rooting,to explore the techniques of Sabina vulgaris cuttage propagation and seedling raising,to find suitable methods for local Sabina vulgaris cuttage propagation,and to promote large-scale seedling production and landscape greening in this area.The means results were as follows:The Sabina vulgaris rooting rate and rooting quality of the cuttings in Tianshan Grand Canyon Scenic area were higher than those in Toli Township.The rooting time of spring cuttage was shorter and the rooting rate was higher and the rooting quality was better.The rooting rate of flat and straight cuttings was higher than that of oblique cuttings,and the rooting condition was better.But it was more favorable for cuttage management and transplanting in the later stage to adopt the method of straight cuttings.The rooting rate and rooting quality of cuttings could be improved in different degree by the combination of the following rooting promoters,concentration and time.Such as,The cuttings were soaked with ABT1 rooting powder with a concentration of 1000 mg·L-1for 2 s or 500 mg·L-1for 1 h,NAA concentration 25 mg·L-1for 4 h or IBA concentration 100 mg·L-1for 1 h.The rooting ability of Sabina vulgaris cuttings was greatly affected by substrate type.In perlite matrix,cuttings have the highest rooting rate and the best rooting quality.The contents of related nutrients,enzyme activities and endogenous hormones in the cuttings were the optimum levels.It was the most suitablesubstrate for cutting propagation of Sabina vulgaris.Rooting characteristics of Sabina vulgaris with difficulty in rooting,low rooting rate and longerrooting time were related to the root system was difficult to penetrate the thick wall tissue,and the cortex rooting was the main way in the rooting type.The physiological changes in cutting process of Sabina vulgaris could be divided into three stages.At the rooting induction stage,The cuttings were in stress because they did not form roots,the contents of soluble sugar and the soluble protein content increased moreover the moisture content and starch content of cuttings decreased at this stage.The activity of SOD,CAT and POD increased in root-induced stage,in order to protected cuttings reduce the adversity stress.The callus and adventitious roots formed gradually in root formation stage during root expression stage.The contents of soluble sugar and the soluble protein content provided at the same time the moisture content and starch content of cuttings increased.Due to root formation reduced stress,so,The activities of SOD and CAT decreased with the decrease of stress.However,the activity of POD reduced first,then accumulation in-Creased,finally reduced and the overall downward trend.At the root growth stage,with the development of adventitious roots in cuttings,the water absorption capacity increased.The soluble protein content and water content of cuttings content increased with the increase of nutrient absorption capacity.But the soluble protein content and starch content decreased continuously.The activities of SOD and CAT decreased,the activity of POD also decreased during the induction period,which was beneficial to cell differentiation and development.During the whole rooting process,GA3and IAA were found to promote the formation and growth of adventitious roots,while ABA was found toas a result of inhibition.The ratio of IAA/ABA was proportional to the rooting rate in the rooting process.During the rooting process,these indexes showed a high degree of coordination with the root development dynamics.
