| Paeonia lactiflora Pall.is a famous traditional flower in China.This research from the domestic main peony varieties selected 11 different colors of peony varieties as experimental material,the peony in full bloom period of design and color with the flower petal structure and the relationship between the environment and the pigment in cells,as well as the five peony varieties to study the design and color with the physiological indexes in the process of flowering.By observing the petals,the organizational structure,the determination of cell sap p H value,moisture content,soluble sugar content,soluble protein content,cyanine total glycosides,total flavones,total carotenoids and condensed tannin content,using high performance liquid chromatography(HPLC)to pigment composition and content determination,comprehensive analysis of the indicators for the influence of the peony design and color,and reveal the mechanism of formation of the peony design and color difference,for the peony design and color genetic research and provides some references for the breeding of new varieties of design and color.The main research results are as follows:(1)The difference of pigment content,composition and synthesis mechanism in petals was the main reason for the difference of flower color in different flowering period.The higher the content of anthocyanin in the petals,the deeper the flower color,the redder and the higher the chroma.The accumulation of Cy3G5 G,Pg3G5G and Pn3G5 G contents all promoted the formation of peony red color.Both Qu3 G and Km3 G show a subtractive effect,which reduces the reddness of the suit.Both Qu7 G and luteolin can improve the yellow color.The increase of carotenoids in petals was the main reason for reducing the redness and promoting the yellow color of peony.Condensed tannin has genotypic characteristics and its accumulation contributes to the formation of dark petals of peony.In addition,the content of condensed tannin was positively correlated with the yellow degree of petals,the total amount of flavonoids and the total amount of carotenoids.(2)The structure of paeonia lanceolata was observed by paraffin section of plant tissue,and the relationship between the anatomical structure and flower color was analyzed.The results showed that with the development of flowering process,the petal area of the tested varieties gradually expanded,and the shape of the epidermal cells changed.(3)The test varieties with higher soluble sugar content had deeper color,and showed reddish purple or purplish red.The variety tested with higher soluble degree content has lighter color.During the flowering process,the soluble sugar content of most samples increased,while the soluble protein content increased first and then decreased.The accumulation of soluble sugar content will reduce the yellow degree of petals.The accumulation of soluble protein content can improve the brightness of petals and reduce the chroma of petals.(4)The cell fluid of the tested cultivar was weakly acidic,and the p H value of the cell fluid was the highest in the yellow petals.In the process of flowering,the p H value of the cell fluid of the pink-white variety increased first and then decreased and then increased.The p H value of cell fluid of the red-purple test variety increased first and then decreased.The variation trend of p H value in cell fluid of yellow and purple cultivars was inconsistent.When the water content of the tested varieties is high,the brightness of the petals can be improved,and the petals can show a shallow color.In the flowering process,the water content of only the powder-white varieties showed the same trend,rising first and then falling.The variation trend of water content of other tested varieties was inconsistent. |
