| In order to explor the effect of spermosphere fungi and endophytic fungi on seed germination and seedling growth of O.henryi.O.henryi seeds and sepermosphere from 3 counties incloud Qinglong Guanling and Sansui in Guizhou province were taken as experimental materials.The fungi collected from different sites were isolated,purified,screened and identified as having the functions of producing IAA,dissolving phosphorus,resolving potassium and fixing nitrogen.Return the fungi promoting solution to the seed,The differences of enzyme activity,endogenous hormone,physiological index and germination index of seeds treated with different fungi at different germination stages were determined and analyzed.Return the fungi promoting solution to sterile sprouts,The differences of growth characters,physiological and biochemical indexes of seedlings treated with different fungi were determined and analyzed.The fungus which can promote seed germination and seedling growth of O.henryi was screened,To provide an ecological and environmental protection method for the efficient reproduction of O.henryi.The results of the study are as following:(1)There were significant differences in germinating index and growth physiological index of palmetto seed between germplasm and sand bed(P<0.05).The indexes of seed germination and physiological growth of seedlings in unsterilized soil bed were better than that in sterilized soil bed.In particular,the root growth index of seedling was significantly higher than that of sand bed and sterilized soil bed.The physiological growth from seed germination to bud,Interseed-microbe play a facilitating role.(2)Isolation,purification,screening and identification of fungi in seeds and seeds were carried out,A total of 17 strains of fungus were isolated,Seven strains of fungi were screened out with the functions of producing IAA,dissolving phosphorus,resolving potassium and fixing nitrogen,which were endophytic fungi QN-3-2(Chaetomium nigricolor)from Qinglong seeds.The spermosphere microorganisms;SS-1-6(Penicillium sp.)SS-2-3(Trichoderma koningiopsis)SS-1-3(Chaunopycnis sp.)QL-3-5(Aureobasidium melanogenum)GL-4-1(Penicillium sclerotiorum)GL-3-1(Mortierella sp.).(3)The indexes of O.henryi seed germination,seed endogenous hormones,related enzyme activity and material metabolism at different germination stages were significantly different under different fungal treatments.The seed germination rate of SS-2-3 was the highest and QN-3-2 was the lowest.The activity of seed-amylase in different treatments changed little with germination time.The decrease of protease after reaching the peak was synchronized with the decrease of seed germination rate.GA3 and ZR increased with different degrees of germination time.The IAA is inconsistent in the variation trend of different treatments.The contents of protein,soluble protein,soluble total sugar and starch in seeds of different treatments all decreased with the extension of germination time.Through principal component analysis and comprehensive evaluation,the three strains with the best cuteness promoting effect were selected.which were GL-3-1(Mortierella sp.)SS-2-3(Trichoderma koningiopsis)and GL-4-1(Penicillium sclerotiorum).(4)The soil diameter,biomass,root index,soluble total sugar,soluble protein and starch of O.henryi seedlings treated with different fungi showed significant differences.However,there was no significant difference in photosynthetic physiological parameters.Through principal component analysis and comprehensive evaluation,it was found that GL-4-1(Penicillium sclerotiorum)SS-1-6(Penicillium sp.)and SS-1-3(Chaunopycnis sp.)had better promoting effects on O.henryi.(5)From the Guanling seed-interstitial fungus GL-4-1(Penicillium sclerotiorum),which is a seed-interstitial microorganism promoting the germination and growth of O.henryi. |
