| Phospholipids are composed of complex systems with different components,fatty acid distribution characteristics and different functions.This paper uses Sunite Sheep brain as the raw material.It aims to understand the composition properties of DHA in sheep brain by determine the content and structure of Sunite Sheep cephalin-bound DHA,and then provides scientific data for the production of new sheep cephalin-bound DHA function food that can prevent and ameliorate brain diseases in the future.First,the crude fat was extracted from 40 Sunite Sheep’s brains’ powder by solvent extraction with the extracted fat by methyl esterification.The fatty acid composition and content of the samples were determined by GC-MS to determine the DHA content included in crude fat of sheep brain.Ice acetone was used in cholesterol removal and phospholipids extraction from crude fat with determination of DHA content in phospholipids.The content of phosphatidylcholine and phosphatidylserine,and the fatty acid composition were further determined by ultraviolet spectrophotometry,reversed-phase high performance liquid chromatography and GC-MS respectively.Then.the fatty acid composition of phospholipids at different sites was determined by lipase decomposition of phospholipids to determine the substructure sites of phospholipid DHA in sheep brain.The results showed that the crude fat of sheep brain extracted by solvent extraction contained 8.89%DHA and the cholesterol content was 29.42±0.92mg/g.It was determined that DHA in the fat of sheep brain accounted for 11.41%of total fatty acids after removing cholesterol.The phosphatidylcholine and phosphatidylserine were 27.67±1.19μg/g and 10.69±1.19±g/g with combined DHA content of 5.58%and 1.72%respectively.The DHA content in the phospholipids greatly increased by lipase decomposition,which indicated that DHA is mainly bound to the Sn-2 position of the phospholipids.The phospholipid DHA extracted from Sunid sheep brain has high nutritional value. |
