In this paper,the leaves,bark,and seed of Acer tegmentosum were used as raw materials.Single factor-Box-Behnken test to determine the best extraction conditions for the volatile oil of A.tegmentosum leaves;Plackett-Burman test-Box-Behnken test was performed on the bark to determine the best extraction conditions for salidroside in the bark;single seed was performed the same experimental design as the leaves.And the essential oil and the oil of seed in three different ways(microwave irradiation,heating sleeve and ultrasonic)were identified by GC-MS,and then calculated the percentage of the essential oil and the oil of seed.The results show:(1)The best conditions for extracting essential oil from leaves in microwave irradiation:the amount of sodium acetate mole is 0.05 mol,the rate of the engine is 450 W,and the time last for 29 min.(2)The best conditions for extracting salidroside of A.tegmentosum bark:liquid-to-material ratio 22.28:1 mL/g,microwave irradiation power 475.36 W,microwave irradiation time 19.38 min.(3)The best conditions for extracting seed oil in microwave irradiation:adding 0.15 g of silicon carbide,16.9 mL/g of liquid-to-material ratio,the rate of the engine is 425 W,and the time last for 11 min,the yield of seed oil is obtained 9.56%.(4)The peaks extracted by the salting-out-microwave irradiation and the peaks extracted by the salting-out-heating mantle reflux are basically the same.The peaks extracted by the microwave irradiation without salt are significantly reduced.A total of 62 compounds were detected by GC-MC in the extraction of essential oil by salting-out and microwave irradiation,54 compounds were detected by GC-MS in the extraction of essential oil by salting-out and reflux extraction by heating sleeve,and 39 compounds were detected by GC-MS in the extraction of essential oil by microwave irradiation without salt.The precipitation of substances in the essential oil would be affected by the absence of salt.The salting-out-microwave irradiation extraction method has many advantages,among which the more prominent are high efficiency,reasonable price and zero degree pollution.It is a technology worth pursuing to extract volatile oil.(5)The seed oil is extracted by microwave irradiation extraction method,heating mantle reflux extraction method,and ultrasonic extraction method.The seed oil components under the three extraction methods all contain palmitic acid,heptadecanoic acid,linoleic acid,and trans-9-octadecenoic acid,stearic acid,erucic acid,behenic acid,etc.The content of linoleic acid and trans-9-octadecenoic acid were the highest under the microwave irradiation extraction method,which were 35.69%and 24.73%respectively.Under the heating mantle reflux extraction method,the content of linoleic acid and erucic acid were the highest,which were 60.67%and 18.94%respectively.Under the ultrasonic extraction method,the contents of linoleic acid and trans-9-octadecenoic acid were the highest,which were 43.6%and 30.26%respectively.Under the three extraction methods,the content of linoleic acid in the three extraction methods is generally higher.The experimental results of this paper not only provide a safe and healthy process route for the extraction and utilization of the essential oil resources of A.tegmentosum,but also provide experimental basis for the application of natural essential oil.
