| Piglet diarrhea is a common disease in pig industry.This experiment was conducted to investigate the preventive and therapeutic effects of Taishan Panshi powder and Rehmannia Glutinosa Polysaccharides(RGP)on piglet diarrhea.The experiment was divided into two parts.Firstly,28 pregnant Kunming female mice were randomly divided into four groups:control group,diarrhea group,Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group,with 7 mice in each group.The mice in the control group and diarrhea group were given normal saline 3.75 m L/kg daily,the mice in the Taishan Panshi powder group were given Taishan Panshi powder Decoction 3.75 m L/kg daily,and the mice in the Rehmannia glutinosa polysaccharide group were given RGP 3.75 m L/kg daily for 5 consecutive days.After delivery,at the age of 13 days,the control group was given 0.4 m L/20 g of normal saline daily and 0.15 m L of sterile normal saline intraperitoneally.The diarrhea group,Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group were given 0.4 m L/20 g of senna leaf Decoction and 0.15 m L of Escherichia coli intraperitoneally for 6 days;The contents of immunoglobulin G(Ig G),immunoglobulin M(Ig M),malondialdehyde(MDA)in serum and the activities of glutathione peroxidase(GSH-Px),superoxide dismutase(SOD),secretory immunoglobulin A(s Ig A)in small intestine and digestive enzyme activities in duodenum,jejunum and ileum were detected by ELISA;The m RNA levels of SGLT1 and NHE3 in jejunum were detected by q-PCR;The expression levels of SGLT1,NHE3,p38MAPK/Ezrin pathway protein and phosphorylation in jejunum were detected by Western blot;He staining was used to observe the development of intestinal morphology.In addition,15 healthy pregnant sows were randomly divided into three groups:control group,Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group,with 5 sows in each group.The control group was given normal basal diet,the Taishan Panshi powder group was given basal diet supplemented with Taishan Panshi powder,75 g/d·head,and the Rehmannia Glutinosa Polysaccharides group was given basal diet supplemented with RGP,75 g/d·head,for 5 consecutive days.The mortality and diarrhea of piglets were observed and detected.The contents of total antioxidant capacity(T-AOC),total superoxide dismutase(T-SOD),glutathione peroxidase(GSH-Px),catalase(CAT),malondialdehyde(MDA),reduced glutathione(GSH)and oxidized glutathione(GSSG)in serum were detected.The results showed that the diarrhea index of the diarrhea group was significantly higher than that of the control group(p<0.05),and the Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group were significantly lower than that of the diarrhea group(p<0.05);Compared with diarrhea group,the villus height and V/C in Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group were significantly increased(p<0.05);Compared with diarrhea group,the activities of SOD and GSH-Px in serum of Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group were significantly increased(p<0.05),and the content of MDA was significantly decreased(p<0.05);The activities of digestive enzymes in duodenum,jejunum and ileum in Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group were significantly higher than those in diarrhea group(p<0.05);Compared with diarrhea group,the expressions of SGLT1 and NHE3 m RNA and protein in Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group were significantly increased(p<0.05),and the phosphorylation level of P38MAPK/Ezrin pathway related protein in Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group was significantly increased(p<0.05).In the sow experiment,compared with the control group,the content of CAT and the activities of SOD and GSH-Px in serum of piglets in Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group were significantly increased(p<0.05),with the highest content at 7 days of age;The contents of T-AOC and GSH in serum of piglets in Taishan Panshi powder group were significantly increased(p<0.05),but there was no significant difference in Rehmannia Glutinosa Polysaccharides group(p>0.05);The contents of MDA and GSSG in serum of piglets in Taishan Panshi powder group and Rehmannia Glutinosa Polysaccharides group were significantly decreased(p<0.05),and the lowest at 21 days of age.Conclusion:Taishan Panshi powder and RGP can reduce diarrhea rate,improve immune function and antioxidant function,increase digestive enzyme activity,and increase intestinal absorption of water,Na~+and nutrients.In addition,Taishan Panshi powder and RGP can improve the antioxidant capacity of piglets,so as to effectively prevent and control piglet diarrhea,and Taishan Panshi powder has the best effect. |
