Objective: To study the change rules of chemical composition in Rehmanniaglutinosa Libosch in different storage conditions, provide a scientific basis for thequality control and storage period of fresh-Rehmannia glutinosa and Rehmanniaglutinosa.Methods: First,to observe and describe the characters of fresh-Rehmanniaglutinosa and Rehmannia glutinosa in the process of storage,with the content ofcatalpol,verbascoside,polysaccharides,monosaccharide and oligosaccharides,aminoacid as a reference index,and determined the moisture,water-soluble extract,totalash,acid insoluble ash,aflatoxin toxins; Determination of catalpol and verbascosidein Rehmannia glutinosa Libosch by HPLC; Monosaccharide and oligosaccharide inRehmannia glutinosa Libosch were determined by the method of RP-HPLC-RID;Polysaccharide in Rehmannia glutinosa Libosch were determined by the method ofphenol-sulfuric;The content of moisture,flooding,ash were measured followingthe Chinese Pharmacopoeia(Part I2010edition);Determination of aflatoxins inRehmannia glutinosa Libosch by HPLC with Post-column Derivatization.Result:1.The study of storage technology for fresh-Rehmannia glutinosa.Use sand,strip boxes,sacks to package the fresh-Rehmannia glutinosa,aftersix months of storage,analysis the influence of fresh-Rehmannia glutinosa indifferent packaging conditions with the detection of the content ofcatalpol,verbascoside,polysaccharides,monosaccharide and oligosaccharides,aminoacid as a reference and determined the moisture,water-soluble extract,total ash,acidinsoluble ash,aflatoxin toxins in fresh-Rehmannia glutinosa. The results showedthat there was a significant difference in moisture content in fresh-Rehmannia glutinosa,the content of moisture in the sand-storage is hige,and the change is little,but the moisture loss fastest is the box-storage,and the fresh-Rehmannia glutinosa of sacks-storage is prone to mildew;Sand-storage was significantly better than the other two packaging methods.Tresh-Rehmannia glutinosa be packed by sand,cool treasury and cellars,compared the effect of storage methods and packaging methods for the quality of fresh-Rehmannia glutinosa.The results showed that after six months of storage,the quality of fresh-Rehmannia glutinosa storaged by cool treasury conditions sand remains the best, content of each component of fresh-Rehmannia glutinosa reduced slowly and the content of moisture changed little,sand storage conditions of fresh-Rehmannia glutinosa,although the moisture content changs little, but after four months of storage,the content of each component decreased rapidly,also,the fresh-Rehmannia glutinosa was sprouting and had moldy phenomenon, fresh-Rehmannia glutinosa storeged with cellaring conditions lost water fastest, and the content of each component changes huge,the quality is unstable.2.The study of storage technology for Rehmannia glutinosaUse the boxes,bags,sacks and PVC material to Rehmannia glutinosa for respectively routine conventional packaging and vacuum packaging,after12months of storage, with the detection of the content of catalpol, verbascoside, polysaccharides, monosaccharide and oligosaccharides, amino acid as a reference and determined the moisture,water-soluble extract,total ash,acid insoluble ash,aflatoxin toxins,inspect the influence of different packaging and storage methods for the quality of Rehmannia glutinosa.The results show that the moisture content of different packaging methods is different largely,the moisture of Rehmannia glutinosa packaged with vacuumed PVC material changed little during storage,so vacuumed PVC is good for the storage of Rehmannia glutinosa,chemical composition in different packaging methods mostly had a downward trend,compared the chemical composition of different package methods,the chemical composition of Rehmannia glutinosa packaged with vacuumed PVC material is the highest of all the package methods,the quality of Rehmannia glutinosa is stable.Packaged the Rehmannia glutinosa with different methods while tests conducted at room temperature,cool experiments and accelerated testing. After storaged for12months,the Rehmannia glutinosa packaged with vacuumed PVC in cool conditions is the best,the content of catalpol, verbascoside, monosaccharide and oligosaccharides,water-soluble extract in Rehmannia glutinosa for other packaged in cool conditions declined slowly,but the moisture increased step by step.At the room temperature,the moisture changed little,but the the content of catalpol, verbascoside, monosaccharide and oligosaccharides,water-soluble extract declined faster under the cool conditions in Rehmannia glutinosa.Conclusion:Understanded the variation of chemical composition of Rehmannia glutinosa and fresh-Rehmannia glutinosa in the storage period.The result of the research provide a scientific basis for the quality control and storage period of Rehmannia glutinosa Libosch.
