Hydrogen-rich Water And Lactulose To Alleviate Ovarian Dysfunction In Weaned Piglets Caused By Moldy Corn

The objective of the present study was to investigate the effect of F.graminearum on the antioxidant levels and NOS of weaned piglets and the detoxification effect of lactulose and hydrogen-rich water on moldy corn.Twenty-four 21-day-old ternary hybrid piglets(du× chang × large)with an average body weight of 7.25±1.02 kg were randomly divided into 4 groups(control group,moldy corn group,lactulose-reducing group and hydrogen-rich water-relieving group).The control group of piglets were fed a normal,non-mildew diet,while the other three groups of piglets were fed a mold-contaminated diet(Deoxynivalenol(DON)and Zearalenone(ZEA)content of 1.1 mg/kg,respectively.0.55 mg/kg).In addition,lactulose and hydrogen-rich water remission groups were administered twice daily with lactulose(500 mg/kg)and hydrogen-rich water(10 ml/kg).The experimental period is 25 days.After 25 days,the piglets were slaughtered,the ovaries were collected and weighed;the morphology of the ovarian tissue of the piglets was observed;the H2,CAT,GSH-PX,T-SOD,SDH,Caspase-3 activities,NO content and NOS activity of ovarian tissues were detected;histochemical localization of nNOS,iNOS,eNOS and CD68.The results of experiment 1 showed that:1)compared with the control group and the moldy corn group,the H2 content in the ovarian tissue was significantly increased(p<0.05),and the H2 content in the lactulose group also increased.2)piglets after eating mildewed corn,the relative weight of piglets’ ovaries was significantly higher than that of the control group(p<0.05),while the relative weight of ovaries decreased after piglets were fed with lactulose and hydrogen-rich water.3)HE results showed that there were no obvious pathological changes in the ovary of the 4 groups of piglets,but compared with the control group,the number of follicles in the piglets fed with moldy corn was significantly increased(p<0.05),the number of follicles follicles growing after lactulose and hydrogen-rich water was decreased compared with the moldy corn group.4)Caspase-3 activity in piglets was significantly increased after feeding moldy corn The activity of Caspase-3 was significantly decreased after lactation with lactulose and hydrogen-rich water(p<0.05).5)immunohistochemistry results showed that the positive rate of CD68 decreased after piglets fed moldy corn,while the positive rate of ovary cells increased after treatment with lactulose and hydrogen-rich water.The results of experiment 2 showed that:1)After feeding moldy corn,the ovarian activity of piglets was significantly increased(p<0.05);the activity of GSH-PX was significantly decreased(p<0.05),and the level of T-SOD was also decreased;Significantly decreased(p<0.05)NO content and total NOS activity increased significantly(p<0.05).2)After cultivating piglets with lactulose,the CAT activity of ovarian tissue decreased,the activity of GSH-PX increased significantly,the level of T-SOD decreased,and the activity of SDH increased significantly(p<0.05);NO level was significantly decreased(p<0.05),and total NOS activity decreased.3)After cultivating piglets with hydrogen-rich water,the CAT activity of ovarian tissue increased with the moldy corn group,the GSH-PX and T-SOD levels decreased,and the SDH activity increased.NO and NOS Activity was significantly reduced(p<0.05).4)Immunohistochemistry results showed that the three NOS(nNOS,iNOS,eNOS)not only expressed in the oocytes of all levels of follicles,but also on granulosa cells and membrane cells.Finally,DON and ZEA in moldy corn may affect the level of NO/NOS,mediate the reduction of antioxidant levels in piglet ovary tissue,and promote mitochondrial function;while lactulose and hydrogen-rich water effectively relieve DON and ZEA by increasing antioxidant levels the harm caused by piglet ovaries.
