| This experiment was aimed to study by alpha lipoic acid,yeast chromium,alpha lipoic acid and yeast chromium mixture of feeding the sheep,the effect of sheep growth performance,nutrient digestion and the influence of liver differential expressed genes under heat stress,this experiment was aimed to research of alpha lipoic acid,yeast chromium on the sheep production performance and the hepatic metabolism of heat stress under the adding dose and add mode.In order to explore the application value provide theoretical basis as feed additives for sheep,and provide reference for optimizing the usage of fodder.Twenty-eight healthy sheep with the similar age and weight were randomly divided into four groups,which were control group(CTL group,fed basic diet),LA group(fed basic diet and alpha-lipoic acid 600 mg/kg,LA group),Cr group(fed basic diet and yeast chromium 0.5 mg/kg,),mixed group(fed basic diet and alpha-lipoic acid 600mg/kg and yeast chromium 0.5mg/kg,MIX group),fed for 50 days.slaughter was carried out at the end of the experiment,and slaughter performance was determined.feces and urine samples were collected,the nutrient digestion and utilization efficiency was determined.To explore the effect of yeast chromium on the immune function of sheep.From the CTL and Cr groups,the liver samples of 3 sheep in each group were selected for high-throughput transcriptome sequencing comparison analysis,and the sequencing results were verified by real-time fluorescent quantitative PCR.The research results are as follows:(1)Dietary yeast chromium supplementation has significantly increased on the carcass weight of sheep under heat stress(P<0.05).(2)Dietary α-lipoic acid and yeast chromium has significantly decreased the contents of the plasma total protein(TP),low density lipoprotein(LDL)and cholesterol(CHOL)(P<0.05).(3)Dietary α-lipoic acid and yeast chromium has significantly increased the blood GH and IL-1β content of sheep under heat stress(P<0.01).(4)Dietary α-lipoic acid has significantly increased plasma IgG content of sheep under heat stress(P<0.01),Cr and MIX groups has significantly decreased plasma IgG content compared with CTL group(P<0.01).(5)Through RNA-Seq(transcriptome sequencing),a total of 821 differentially expressed genes were found,of which 341 were up-regulated and 480 were down-regulated.The qRT-PCR results of 8 differentially expressed genes are consistent with the sequencing results.(6)The expression levels of immunorelated genes were analyzed by fluorescence quantitative data,It was found that yeast chromium up-regulate the expression of CD80 gene,and down-regulate the expression of CD86.(7)Yeast chromium supplementation in the diet can protect sheep from liver damage.In conclusion,adding alpha-lipoic acid and yeast chromium to the diet can improve the immunity and antioxidant capacity of the body,yeast chromium can promote animal feeding,increase carcass weight,and also play a certain protective effect on the liver. |
