Persimmon has good ecological significance and economic value.Its fruit persimmon is rich in nutrition and is one of the important fruits.China is the main persimmon producing country,however,the occurrence and prevalence of persimmon anthracnose has brought a fatal blow to persimmon planting industry.The results showed that the microbial community structure was closely related to the occurrence of plant diseases.At present,researches on persimmon anthracnose mainly focus on pathogen identification and chemical control of the disease,but there is no report on the relationship between microbial community structure and persimmon anthracnose.In this paper,two persimmon plantations in Fangshan city of Yongkang,Zhejiang Province,which are "the hometown of Persimmon in Fangshan of China",were selected for ecological management and over management.The samples were collected in summer and autumn,and the total DNA of soil and branches were extracted In this study,the relationship between the occurrence of persimmon anthracnose and the changes of endophytic microbial community structure in the soil around the root and branches of persimmon was studied to provide a theoretical basis for the biological control of persimmon anthracnose.The main results are as follows:1.The results of high-throughput analysis showed that at the genus level,the dominant species of bacteria in the soil around the roots of persimmon trees in Fangshan city were Bacillus,Burkholderia-caballeronia-paraburkholderia,Arthrobacter,Acidothermus and Alicyclobacillus.The dominant species of fungi were Talaromyces,Saitozyma,Fusarium,Cylindrocarpon and Apiotrichum.The dominant species of endophytic bacteria in persimmon branches with different degrees of disease were Erwinia,Curtobacterium,Burkholderia-caballeronia-paraburkholderia and Methylobacterium-methylumbrum were identified.The order of population diversity was soil bacteria >soil fungi > endophytic bacteria in branches.2.In the ecological management area,the diversity of bacteria and fungi in the soil around the root of persimmon trees increased with the increase of the incidence of anthracnose,and the infection of persimmon anthracnose led to the increase of the diversity of endophytic bacteria in the diseased branches.In the over management area,with the increase of the incidence of anthracnose,the diversity of bacteria showed an upward trend,but the diversity of fungi showed a downward trend.With the development of persimmon anthracnose,the diversity of endophytic bacteria in persimmon branches showed an upward trend.3.In the same season,the relative abundances of Bacillus,Bacillus lipocalis,Bacillus polymyxa,Sarcococcus and Burkholderia in the soil around the roots of persimmon trees with different disease degrees in the ecological management area were higher than those in the excessive management area.With the increase of the incidence of persimmon anthracnose,the relative abundance of Bacillus,Bacillus lipocalis,Bacillus polymyxa and Sarcococcus in the soil around the root of persimmon decreased,while the relative abundance of Burkholderia increased.4.The relative abundance of Aspergillus flavus in the soil around the root of Persimmon in the ecological management area was higher than that in the over management area.With the increase of the incidence of persimmon anthracnose,the relative abundance of the Saitozyma in the soil around the root of persimmon increased.5.In the ecological management area and over management area,the relative abundances of two endophytic bacterial groups of Picobacteria and Methylbacilli in diseased branches of seriously diseased persimmon plants were generally higher than those in healthy branches,that is,the abundances of the above two bacterial groups showed an increasing trend in severely diseased branches.There was no significant correlation between the changes of other endophytic bacteria and persimmon anthracnose.In conclusion,The diversity of microbial flora in the soil around the roots of the transition zone is higher.In the soil around the root of persimmon,the relative abundance of Bacillus,Bacillus lipocalis,Bacillus polymyxa,and Sarcococcus decreased with the increase of the degree of anthracnose,while the relative abundance of Burkholderia increased with the increase of the degree of anthracnose.In the soil around the root of persimmon,the relative abundance of Saitozyma increased with the increase of the incidence of persimmon anthracnose.In the branches of persimmon trees,the relative abundance of Bacteroides and methylbacteroides increased with the increase of the degree of anthracnose.
