| Ujumqin sheep and Hu sheep,as two excellent local sheep breeds in China,has good meat performance,but lack the corresponding basic research in molecular genetic breeding.Therefore,DGAT1,FUT8,PTPRG,MAN1A1,GHR,PFKFB4 and NR6A1 genes related to animal growth and development were selected as candidate genes.In this study,the SNP loci of DGAT1,FUT8,PTPRG,MAN1A1,GHR,PFKFB4 and NR6A1 genes were genotyped in 336 Ujumqin sheep in Inner Mongolia and 1006 Hu sheep in Gansu.The genotyping of the SNP loci of DGAT1,FUT8,PTPRG,MAN1A1,GHR,PFKFB4 and NR6A1 genes was carried out by Sequenom Mass ARRAY~?SNP technology and gene chip.The genetic information of the population related to each SNP locus was statistically analyzed,and the correlation analysis was carried out between SNPs and growth traits of Ujumqin sheep.The results of the study are as follows:1.In this study,the genotype detection and analysis of 7 SNPs genes found that these 7 loci were polymorphic and belong to moderate polymorphic(0.25<PIC<0.5)in the Ujumqin sheep population.In the Hu sheep population,g.51303722A>G locus belongs to low polymorphism(PIC<0.25),and g.18795097C>T locus belongs to high polymorphism(PIC>0.5).Chi-square test showed that the 7SNPs in the Ujumqin sheep population all reached Hardy-Weinberg equilibrium(P>0.05).Among them,g.18795097C>T and g.51303722A>G also reached Hardy-Weinberg equilibrium in the Hu sheep population(P>0.05).2.Association analysis of genetic polymorphisms and population growth traits of Ujumqin sheep revealed that at the age of 4 months and 6 months,the TT,CC and CT genotype at the g.13504098C>T locus of DGAT1 gene showed significant differences in chest girth,body weight,shin circumference,body height and chest width(P<0.05);the TC and CC genotype at the g.74522417C>T locus of FUT8gene showed significant differences in body height(P<0.05);the GG,AA and GA genotype at the g.39335737A>G locus of PTPRG gene showed significant differences in shin circumference,chest girth,bod weight and body height(P<0.05);the CC,TC and TT genotype at the g.18795097C>T locus of MAN1A1 gene showed significant differences in chest girth,chest width,body weight and body height(P<0.05);the CC,TT and TC genotype at the g.31957140C>T locus of GHR gene showed significant differences in body height and chest width(P<0.05);the GG,GA and AA genotype at the g.51303722A>G locus of PFKFB4 gene showed significant differences in shin circumference,chest girth and body weight(P<0.05);the CC,CA and AA genotype at the g.11204707A>C locus of NR6A1gene showed significant differences in body weight,body height,body length,chest girth and shin circumference(P<0.05).3.Association analysis of genetic polymorphisms and population growth traits of Hu sheep revealed that at the age of 2,3,4,5 and 6 months,the CC,TC and TT genotype at the g.18795097C>T locus of MAN1A1 gene showed significant differences in body weight,chest width,back fat thickness,eye muscle area,chest depth,body length(P<0.05);the differences in body length,chest width,between the GG,GA and AA genotype at the g.51303722A>G polymorphic locus of PFKFB4 gene at 4months of age were significant difference(P<0.05).The results of this study are of great significance for further understanding and mastering the growth and development of Ujumqin sheep and Hu sheep,screening and verifying SNP sites that affect their growth traits,which are of great significance for better utilization of breed effects,and are also molecular Marker-assisted selection breeding provides a reference. |
