O.biennis L.stems have good anti-inflammatory and antioxidant activities because its rich in flavonoids.However,the optimal extraction conditions for flavonoids in O.biennis L.stems and seeds are still unclear.This study uses total flavonoids as a reference index for the first time,and uses flash extraction technology and response surface analysis to optimize the extraction conditions of O.biennis L.stems(OBSE)and seeds(OBSD).At the same time,because there are few reports on the antibacterial activity of O.biennis L.,in order to further explore the antibacterial activity and antibacterial mechanism,this study takes S.aureus as the object to discuss the antibacterial effect and mechanism of O.biennis L.from cell permeability and respiratory metabolism.The results of antibacterial experiments show that OBSE and OBSD have a good inhibitory effect on S.aureus.The diameters of the inhibition zone were 13.75 mm and 12.70 mm,and the minimum inhibitory concentrations were 1mg/m L and 2 mg/m L,respectively.After OBSE and OBSD treatment,the bacterial contents leaked,the alkaline phosphatase activity,conductivity,nucleic acid and protein levels in the bacterial suspension were significantly reduced(p<0.01),it was also observed that the surface of S.aureus had morphological changes such as cracks and shrinkage by scanning electron microscope.Through the detection of the respiratory inhibition rate and the superposition rate,it is determined that OBSE can affect the respiratory metabolism of S.aureus through the pentose phosphate pathway,and OBSD can affect the respiratory metabolism of S.aureus through the glycolytic pathway.The key enzymes glucose-6-phosphate dehydrogenase,phosphofructokinase,protein kinase and hexokinase of the corresponding pathways were tested,and their contents were significantly reduced(p<0.01).In summary,both OBSE and OBSD have an inhibitory effect on S.aureus by affecting the permeability of bacterial membranes and respiratory metabolism.
