| This experiment was conducted to study the effects of different dosage forms of sodium butyrate on production performance,egg quality,serum biochemical indexes,intestinal microflora,immune and intestinal barrier related gene expression levels of laying ducks.To explore the effects of sodium butyrate on promoting production,improving immune function and intestinal health,and screening out the appropriate dosage form and additive dose to provide a reference for the application of sodium butyrate in laying duck production practice.A total of 840 48 week-old Shendan 2 laying ducks were randomly allotted to 7 treatments with six replicates,with 20 ducks in each replicate.The group Ⅰ was the basal diet group,while Ⅱ,Ⅲ,Ⅳ,Ⅴ,Ⅵ,Ⅶ group as the experimental group.The control group was fed the basal diet,while the experimental group was fed the basal diet with 500 g/t,1 000 g/t,2 000 g/t granulated sodium butyrate and 250 g/t,500 g/t,1 000 g/t coated sodium butyrate,respectively.After 8weeks of feeding with sodium butyrate,the results showed that:1)Adding sodium butyrate can improve the performance of laying ducks.Compared with group Ⅰ,group Ⅴ significantly increased the egg production(P<0.01)and feed conversion rate(P<0.05)of laying ducks in 53 to 56 weeks.Compared with group III,group V significantly improved egg-laying and feed conversion efficiency(P<0.01)in 53 to 56 weeks.In terms of the quality of eggs,compared with group Ⅰ,group Ⅴ,group Ⅵ and group Ⅶ were significantly improved the duck egg weight(P<0.05),group Ⅳ significantly improved the duck egg weight(P<0.01).What is more,the proportion of egg yolk in group V,VI and VII was significantly higher than that in group IV(P<0.05),and egg yellow was significantly higher in group V and group VII than in group IV(P<0.05)2)Diets supplemented with different proportion and dosage forms of sodium butyrate can increase the contents of Ig G,Ig A and Ig M in serum.Compared with group Ⅰ,group Ⅴ and group Ⅵsignificantly increased the Ig G(P<0.05)and Ig M(P<0.01)content,and group Ⅶ can improve the Ig G(P<0.01),Ig A(P<0.05)and Ig M(P<0.05)content,while group Ⅱ can significantly improve the Ig M(P<0.01),the Ig A(P<0.05)and Ig G levels(P <0.05).Compared with group I,T-AOC、SOD and CAT levels in the serum of laying ducks were increased at different levels.And group V,VI,VII significantly decreased the MDA content compared with group I and group II(P<0.05).Comparison between Ⅴ,Ⅵand Ⅶ groups,T-AOC contents in group V and group VII were significantly higher than those in group VI(P<0.05).3)Dietary sodium butyrate decreased the content of inflammatory factors in serum.Compared with group Ⅰ,group Ⅱ,Ⅴ,Ⅵ,Ⅶ have significantly reduced the TNF-α and γ-IFN content(P<0.01).Besides the IL-1β content in all experimental groups was significantly decreased(P<0.01)and group Ⅵsignificantly reduced the content of IL-6(P<0.01).Group Ⅶ was significantly below compared with group Ⅲ and Ⅳ(P<0.01),the contents of IL-6 in group Ⅵ was significantly below compared with group Ⅲ(P<0.05).The addition of different dosage forms of sodium butyrate decreased the expression of IL-1β,IL-6,γ-IFN and TNF-α in the spleen tissue to varying degrees,in which compared with groupⅠ,group Ⅱ,Ⅳ,Ⅶ significantly down-regulated the expression of IL-1β(P<0.05),and group Ⅶdown-regulated the expression of TNF-α(P<0.05).There was no significant difference between the two dosage forms(P>0.05).4)Different dosage forms of sodium butyrate could increase the gene expression of ZO-1,Occludin and Claudin-1 in ileum of laying ducks,among which significantly raised the expression of Claudin-1m RNA in group Ⅶ(P<0.05),and the expression of ileal Occludin m RNA were significantly increased inⅣ and Ⅴ group(P<0.05).The effect of two dosage forms of sodium butyrate was basically the same.5)Dietary sodium butyrate can increase the Alpha diversity of ileum and cecum microorganisms.Group Ⅱ significantly increased the ileum microorganism Chao1 index(P<0.05).Adding sodium butyrate can optimize the intestinal microflora and increase the proportion of Firmicutes in ileum and cecum at the phylum level and increase the proportion of Lactobacillus in ileum at the genus level.In conclusion,dietary sodium butyrate can improve the performance of immune function and antioxidant capacity,stimulate the expression of intestinal barrier related genes,regulate intestinal microflora,and ultimately improve intestinal and body health of laying ducks.Under experiment’s conditions,the coated sodium butyrate group is overall effect were better than that in the granulated sodium butyrate group,and 250 g/t coated sodium butyrate was the optimal supplemental amount. |
