| Chinese Cabbage(Brassica campestris ssp pekinensis),originated in China,is one of the important vegetable crops in China.The flower colors of Chinese cabbage are mainly yellow,orange and white,which are not only easy to observe,but also stable inheritance.In the process of production practice and plant resource conservation,flower color can be used to identify whether the plant is homozygous,thus eliminating hybrid plants to improve the purity of parents;flower color can also be used to calculate the hybridization rate,so as to test the purity of hybrid.Therefore,the study on the genetic law of flower color of Chinese cabbage can accelerate the process of Chinese cabbage cross breeding.At the same time,it laid a foundation for further elucidating the mechanism of flower color variation in Chinese cabbage.1.In this study,the pure lines of Chinese cabbage with yellow,orange and white flower colors were selected as the test materials,and F1,F2,F3,BC1,triple cross and other generation materials were obtained by means of cross,self cross and backcross.Flower color identification was conducted at flowering stage,and the data were statistically analyzed.The results showed that all F1 progenies of reciprocal cross between orange flower,yellow flower and white flower were yellow plants,and the characters of F1 selfing progenies were separated,and the relationship between yellow flower and orange flower and white flower was obvious and recessive controlled by single gene.However,it is difficult to explain the segregation of flower color into yellow,orange and white in F2 generation after cross between orange flower and white flower.Therefore,it is concluded that the inheritance of yellow flower,white flower and orange flower color in Chinese cabbage used in this study is an interactive inheritance regulated by recessive epistatic genes.2.The recessive epistatic gene in this experiment means that A and a can perform their respective roles when the basic color gene B exists,that is,the yellow flower gene is A_;The orange flower gene was aa;In the absence of B gene,recessive gene B plays an epistatic role on A、a,which makes A、a unable to show its role,thus showing white flower.3.In order to further study the mechanism of flower color variation,this study also carried out metabonomics analysis on yellow flowers and white flowers,and found Lutein and(E/Z)-Phytoene in carotenoids,Among the flavonoids,11 different metabolites were identified as Kaempferol-3-O-(2’’-acetyl)glucoside*and 5,4’-Dihydroxy-7-methoxyflavanone(Sakuranetin)*,And the four substances that change in the synthesis of flavonoids:Tricetin,Homoeriodictyol,Apigenin and Sakura,the expression difference between the two colors is extremely significant,so it is speculated that the changes of the two colors may be related to the pigment types such as carotenoids and flavonoids or their content changes. |
