| The lung is the most important organ of the respiratory system.When broilers suffer from lung injury,it will cause pulmonary edema,alveolar epithelial cell damage,release a large number of inflammatory cells and inflammatory factors,and cause the death of broilers in severe cases and cause serious economic losses to the breeding industry.Taurine can inhibit the inflammatory response induced by multiple tissues and organs of the animal body and regulate the TLR4/NF-κB signaling pathway.However,whether taurine can protect broilers from lung injury by alleviating pulmonary edema and inhibiting inflammation is still unclear.This study will establish a broiler lung injury model by intraperitoneal injection of lipopolysaccharide(LPS)in broilers,and add taurine to the diet preventively to explore the effect of taurine on lung injury in broilers.Provide theoretical basis in the application of alleviating lung inflammation and preventing lung injury.In the experiment,120 white-feather broilers at the age of 1 day were selected and divided into 4 treatments,each treatment was divided into 6 replicates and 5 replicates each.The blank control group(C)and model group(M)were fed with basal diets.The taurine control group(T)and the taurine prevention group(MT)were supplemented with 0.3% taurine in the basal diet.The blank control group(C)and the taurine control group(T)were used on the 26 th,27th,and 27 th days.An equal volume of saline was injected intraperitoneally on 28 days;the model group(M)and the taurine prevention group(MT)were injected intraperitoneally with LPS(1 mg/kg.BW)on days 26,27,and 28.Apply HE staining,enzyme-linked immunosorbent assay(ELISA),real-time fluorescent quantitative PCR technology and western blotting techniques,To study the effect of preventive addition of taurine on LPS/TLR4/NF-κB signaling pathway and inflammasome NLRP3 in broiler lungs induced by LPS.The test results showed that taurine significantly reduced pulmonary edema and MPO activity in lung tissue of broilers with lung injury induced by LPS(P<0.01);Taurine reduced the total protein concentration in broiler lung lavage fluid(BALF)(P<0.05)in lung injury induced by LPS(P<0.05),and significantly reduced the content of white blood cells,macrophages,and neutrophils in BALF(P<0.01),Reduce the content of lymphocytes(P<0.05);In terms of inflammatory factors,taurine extremely significantly reduced the levels of IL-1β,IL-8,IL-6,TNF-α,IL-12,IFN-γ in serum and lung tissue of broiler chickens with lung injury induced by LPS(P<0.01),while extremely significantly increasing the content of IL-4 and IL-10(P<0.01);Taurine significantly reduced the expression levels of TLR4 and NF-κB genes and proteins in the lung tissues of broilers with lung injury induced by LPS(P<0.01)and reduced the expression levels of TRAF6,My D88,and NLRP3 m RNA to varying degrees.At the same time,the expression levels of IκB and PPAR-γ m RNA were increased to varying degrees.To sum up,taurine can alleviate pulmonary edema and inhibit the excessive secretion of inflammatory mediators in broilers with lung injury and the activity of MPO in lung tissues.Through PPAR-γ activation,it can inhibit LPS/TLR4/NF-κB signaling pathway to down-regulate NLRP3.Thereby alleviating lung inflammation,reducing lung damage,and protecting the lungs.. |
