| A lot of variation phenomena has been found in the industrialization of jujube in southern Xinjiang,which is manifested by the differences in fruit shape,fruit weight,fruit flavor and internal quality in terms of fruit traits.This study systematically investigated the botanical and biological traits of excellent strains of the bud mutation ‘Huizao’ jujube which found during in the production,and comprehensively evaluated their fruit economic traits.At the same tine the excellent strains also have been combined with SCoT molecular marker technology to identify differences between ‘Huizao’ jujube;Aim to provide reference for the early screening and identification of new varietal varieties,to screen out new lines with good quality,and to reserve materials for the selection and breeding of good varieties.The main research results are as follows:1.In the phytomorphology of each excellent strains material,there are certain differences in leaves,branches,spines,hangings and fruits,and the variation range is 9.18%~79.74%.The differences between the excellent strains and the ‘Huizao’ jujube could be identified from the phytomorphology.2.Each excellent strains material had some differences in fruit quality,and a comprehensive evaluation of fruit economic traits was conducted by applying principal component analysis,and the evaluation results were as follows.Fruit freshness: from high to low,ZB-1,DH-1,Y-36,‘Huizao’ jujube,9-1,ST-CK,and ST-2,respectively.Dried fruits: ZB-1,ST-CK,ST-2,DH-1,9-1,‘Huizao’ jujube,Y-36 from high to low.3.The differences among the strain were further confirmed by applying SCoT molecular marker technology.8 primers with good polymorphism were selected from 78 primers,and a total of 98 clear and reproducible bands were amplified,with bands distributed from 100 to 2000 bp.12.25 bands were amplified for each primer on average,including 52 polymorphic bands,with an average polymorphism percentage of 45.97%.Primer 42 amplified the largest number of bands,24,while primer 43 had the highest polymorphism percentage of 81.81% and primer 59 had the lowest percentage of 18.18%.The differences in the DNA level among the test materials could be seen from the SCoT molecular markers,which are new strains distinguishing from the ‘Huizao’ jujube. |
