| Rabbit hemorrhagic disease is a highly infectious and extremely fatal infectious disease(90%~100%)caused by rabbit hemorrhagic disease virus(RHDV).The virus belongs to the family calicivirigae.Since it was first reported in Jiangsu,China in 1984,it has been gradually reported in various countries and regions around the world,resulting in the death of tens of millions of wild and domestic adult rabbits.It has become a common and devastating infectious disease.At the same time,it also threatens whether the whole rabbit population can be preserved by nature.RHDV mainly invades the lung,liver,kidney,heart and spleen of rabbits,especially the liver.The liver is the main site of RHDV proliferation.Through the proteomics study of the liver,the pathogenic mechanism of why young rabbits are not easy to be infected and adult rabbits are easy to be infected in the process of RHDV infection is studied.In addition,the systemic immune system is also affected by the spread of the virus.Therefore,it is also of great significance to study the role of some proinflammatory factors in peripheral blood leukocytes(PBL)in the pathogenesis of RHD.The aim of this study was to evaluate the changes of nine proinflammatory cytokines(IL-6,IL-8,IL-10,IL-12,IL-17,IFN-γ,TNF-α,TGF-β and MCP-1)in peripheral blood leukocytes of adult rabbits artificially infected with RHDV as experimental group and adult rabbits not infected with RHDV as control group by ELISA.The expressions of IL-8,IL-10,MCP-1,IFN-γ and TNF-α were significantly up-regulated,but there was no significant change in IL-6,IL-12,IL-17 and TGF-β.It is clarified that the changes of IL-8,IL-10,MCP-1,IFN-γ and TNF-α in the cytokines studied have certain influence on the survival time of infected rabbits.TMT quantitative proteomics was used to analyze the protein expression in the liver of uninfected adult rabbits and young rabbits.821 differentially expressed proteins were obtained,of which 294 were significantly up-regulated and 527 were significantly down regulated.Bioinformatics analysis showed that these proteins were mainly related to metabolic pathways and substance transport,and some of them belonged to structural proteins and other proteins.It laid a foundation for further study on the pathogenesis of young and adult rabbits. |
