Characteristics Of Marigold ’Xueyu No.3’ Variety And Construction Of DNA Fingerprint

Abstract/Summary:
The breeding process and characteristics of marigold ’Xueyu No.3’ were described in this study.Using ’Xueyu No.3’ and 5 control marigold varieties as experimental materials,according to the DUS test guidelines of marigold genus,the differences,clustering,correlation and principal components of the materials were analyzed by using 23 character indexes,and the ornamental value was evaluated.The DNA fingerprints of ’Xueyu 3’ and its five control varieties were constructed based on SSR molecular markers.This study provided an important theoretical basis for enriching genetic resources of marigold and new varieties for ornamental use in Qinghai.The results are as follows:(1)In 2012,the Plateau Flower Research Center of Qinghai University used C40-1(sterile plants)as the female parent and C-11(fertile plants)as the male parent,and configured the hybrid combination C40-1×C-11.From 2013 to 2017Multi-generation single plant and group selection to form a new line.Variety comparison trials,regional trials and production trials were conducted from 2018 to2020.In 2020,it was identified and put on record as a new variety by Qinghai Province Crop New Variety Examination Committee,and named Marigold ‘Xueyu No.3’.(2)According to the DUS detection guidelines of marigold,the coefficient of variation and correlation of 23 phenotypic traits were analyzed.The results showed that the highest coefficient of variation was anthocyanin chromogenic in capitulum pedicel.The branching was correlated with the length of liguulate floret,the width of terminal leaflet and the notch depth of leaf margin.Cluster analysis showed that the six cultivars were divided into three classes when the Euclidean distance was 10.Principal component analysis showed that the cumulative contribution rate of the four principal components reached 98.14%.The comprehensive evaluation results show that ’Xueyu No.3’ has the highest ornamental value.(3)Four pairs of primers were used for SSR-PCR amplification of the experimental material.The primers PIC was 0.355,Shannon’s index was 0.592,and Nei’s gene diversity was 0.407.UPGMA cluster analysis showed that the 6 marigold varieties were divided into 4 groups at the similarity coefficient of 0.744.Four pairs of primers were used to construct the DNA fingerprints of ’Xueyu No.3’ and its five control varieties.
