| Grassland black fungi is a kind of new edible fungus,which has domesticated from wild fungus in recent years,with a high nutritional value,nutritional value and medicinal value.It is rich in protein,vitamins,amino acids,carbohydrates and calcium,phosphorus,iron,zinc and other trace elements essential to human body.However,Grassland black fungi is sensitive to the external environment and grows slowly under natural conditions.In order to provide good strains for artificial cultivation,we used the protoplasts of Grassland black fungi as the starting material for UV mutagenesis,thus to improve the growth rate of Grassland black fungi’s mycelium,and to provide the basis for give higher yield and more nutritional value.The main conclusions of this experiment are as follows:1.Using the mycelium of hm8124-1 and hm910 as the material with liquid shaking for 8 days,the concentration of lysozyme was 1.4%,and the time of enzymolysis was 4 hours at 25℃,could be enough for the preparation of protoplasts of Grassland black fungi.2.The protoplasts of the two strains hm8124-1 and hm910 were irradiated with ultraviolet radiation for different time,and the mortality increased with the extension of ultraviolet radiation time,but decreased with the increase of distance from UV lamp.By which we infer the fatality rate is proportional to mutagenesis time and inversely proportional to mutagenesis distance.3.After mutagenesis,single colonies with fast growth and strong growth were selected to measure the mycelium growth rate and extracellular enzyme activity.The materials with faster growth rate were selected from the regenerated strains,and the activities of laccase and protease were changed in the regenerated strains.4.After ISSR analysis of the regenerated mycelium,there were significant differences in the electrophoresis patterns of PCR products between the mutated strain and the control strain,which indicating that the DNA sequence had changed. |
