| In this paper,BYT,M82,LCYB2 gene-edited tomato(L2)and CYCB gene-edited tomato(CB)were used as experimental materials.The HPLC conditions for the determination of lycopene content were optimized.The content of pigment in BYT fruits at different ripening stages and 4 different types of red ripening stage and the expression levels of 7 key lycopene metabolism genes(PSY1,PSY2,PDS,ZDS,LCYB1,LCYB2 and LCYE)in green,white,pink and red ripening stage of BYT fruits were analyzed.The main results are as follows:(1)By optimizing the HPLC conditions for the determination of lycopene content,a rapid quantitative analysis method for lycopene was established.(2)During the ripening process of BYT fruit,the content of lycopene and phytoene increased gradually,and reached the maximum value at the red ripening stage.(3)The total contents of lycopene and phytoene in the fruits of 4 different types of lycopene at ripening stage were arranged in order by size: L2>CB> BYT>M82.The results showed that knockdown of LCYB2,a key gene downstream of lycopene metabolism pathway,could significantly increase the content of lycopene and phytoene.(4)The expression levels of seven key lycopene anabolic genes were different at different stages of BYT fruit development.The results showed that PSY1 gene,LCYB2 gene and LCYE gene were involved in lycopene accumulation and regulation of fruit color change in BYT fruit. |
