F-2 toxin,also known as zearalenone(Zearalenone),because of its estrogen-like effect can cause hyperestrogen in livestock and poultry and interfere with related reproductive performance.Tannic acid is a polyphenol substance widely present in nature.Its special pharmacological activity and structural multiplicity make it have anti-inflammatory,anti-viral and anti-tumor properties.Objective:In this experiment,Kunming mice were used as the research object to explore the protective effect and possible mechanism of tannic acid on F-2 toxin-induced ovarian injury in mice,so as to develop tannic acid as an antidote for F-2 toxin Provide experimental basis.Methods:90 healthy 7w Kunming female rats were randomly divided into 6 groups,each group has 15 rats,namely control group,F-2 toxin group,F-2 toxin + low,medium and high tannic acid group(50,100,200 mg / kg · bw)and F-2 toxin + 100 mg / kg · bw vitamin E group.The control group was infused with 10% ethanol solution,and the other groups were infused with F-2 toxin suspension(10mg / kg,dissolved in 10% ethanol),once per day for 7 consecutive days to induce a reproductive system injury model;The control group and the F-2 toxin group were given double-distilled water,and the remaining groups were given the corresponding doses of tannic acid and vitamin E once a day for 7 days.Subsequently,the mice were sacrificed,and blood and reproductive organs were collected for the following experiments: detection of reproductive hormones Estradiol(E2),Follicle Stimulating Hormone(FSH),and Luteinizing Hormone(LH)levels in the serum of mice;Detection of ovarian reactive oxygen species(ROS)levels,superoxide dismutase(Superoxide dismutase,SOD),glutathione(Glutathione,GSH),malondialdehyde(Malondialdehyde,MDA)levels;H & E staining Observe the morphological changes of ovary,Tunel method to detect ovarian cell apoptosis;immunohistochemical method to detect the expression of caspase-3,caspase-8,caspase-9,estrogen receptor(ER)in mouse ovary;Western blot was used to detect the expression of Fas,Fas L,caspase-8,and mitochondrial apoptosis signaling pathways Bax,Bcl-2,caspase-3,and caspase-9.Results:1)The tannic acid treatment group can effectively relieve the appearance of ovarian congestion and redness caused by F-2 toxin,and significantly reduce the ovarian organ coefficient(P <0.05);2)After tannic acid treatment,F-2 toxin caused The estrous cycle disorder of mice was restored to normal;3)100mg / kg and 200 mg / kg tannic acid can significantly reduce the increase of reproductive hormone E2 level caused by F-2 toxin(P<0.05);compared with the control group,Although F-2 toxin can reduce the level of LH and FSH in the body,the difference is not significant;compared with the F-2 toxin group after adding tannic acid,only 200 mg / kg tannic acid can significantly reduce the LH and FSH Levels(P <0.05);and there was no significant difference in LH / FSH between the groups;4)H & E staining of the ovary found that compared with the F-2 toxin group,tannic acid significantly increased the number of healthy growing follicles;5)There was no significant difference in SOD activity among the experimental groups;compared with the control group,the CAT activity of the F-2 toxin group was significantly increased(P <0.05).After adding tannic acid,the CAT activity was further increased compared with the F-2 toxin group,of which 100 mg / kg and 200 mg / kg tannic acid group was significantly different(P <0.01);compared with the control group,F-2 poison The GSH content of the group was significantly reduced(P <0.05).Compared with the F-2 toxin group after adding tannic acid,the GSH content of the 50 mg / kg tannic acid group decreased significantly(P <0.01),while100 mg / kg and 200 mg / However,the GSH content in the kg tannic acid group increased to a certain extent,and the difference was not significant;the addition of tannic acid can significantly reduce the increase of ovarian ROS and MDA levels caused by F-2 toxin(P<0.01);6)The addition of tannic acid can significantly inhibit the apoptosis of ovarian granulosa cells induced by F-2 toxin;7)The results of immunohistochemistry show that200 mg / kg tannic acid can significantly and significantly reduce the ovary caused by F-2toxin caspase-3,caspase-8,caspase-9 and ER expression increased;8)Western blot detection found that tannic acid treatment group can reduce Fas,Fas L,caspase-8,caspase-3 caused by F-2 toxin And caspase-9 expression and Bax / Bcl-2 increased(P <0.01).Conclusion:Tannic acid has a repairing effect on ovarian injury caused by F-2.It can improve the antioxidant capacity of reproductive organs,remove ROS levels in the ovary,reduce the apoptosis rate of ovarian granulosa cells,and restore related reproductive hormone levels and clearing cycle In order to achieve the repair effect;this repair effect may be closely related to the inhibition of ER and ROS-mediated death receptors and mitochondrial apoptosis signaling pathways by tannic acid.
