| The black bear,known as the Black Blind bear,is a mammal of the genus Ursus in the order Carnivora.This study carried out the rescue and breeding of the black bears,which successfully protected 39 adult Black Bears from casualties and breed five Black Bear cubs,would be the foundation for the scientific protection of black bears.The study,including the design and management of black bear’s cage,the rules of feeding safety,the rules of feeding staff management;the optimization of feeding formula,the daily behavior of black bear;the regular of breeding were studied,;How to rescue Black Bears;disease control and clinical case studies to find ways to treat Black Bears;genetic sequence analysis of Ascaris lumbricoides,the most common large parasite of black bears,to identify ascaris species,provides the theoretical basis for the Black Bear to deworm.In this study,the cytochrome I cox1 gene was amplified by PCR from mitochondrial DNA of four Ascaris Black Bear samples collected from Changsha ecologic zoo and Xiyou Animal Breeding Center in Tianxin District,Changsha.DNA analysis software DNAStar 5.0was used to compare the sequence with that of ascaris in Gen Bank and in other wildlife.The results showed that the lengths of cox1 of Ascaris from Changsha ecologic zoo and Xiyou Animal Breeding Center were 400 bp to 414bp;Compared these sequences of Ascaris gene with those of non-native Ascaris populations: the highest homology was 96.3% to 97.5%with the Netherlands Baylisascaris transfuga strain KC543477.1;phylogenetic tree revealed that the 4 Hunan isolates were clustered into the same clade of the Baylisascaris transfuga reference strains available in Gen Bank.As the results,Black bear ascaris and Baylisascaris transfuga had the highest homology with each other,and the sequence of Pcox1 was relatively conservative in this study. |
