| Potato(Solanum tuberosum L.)is the fourth largest food crop and the largest crop for food and vegetable in China.Inner Mongolia is one of the main potato production areas in China,but the shortage of specialized varieties has become a key problem restricting the independent innovation and competitiveness of the potato seed industry.In this study,the morphological characteristics,yield,nutritional quality and other traits of 18 parents and their hybrid offspring were observed,and the genetic differences among the parents and the authenticity of the hybrid offspring were identified with SSR molecular marker technology.It would lay a foundation for the breeding of new potato varieties.The main results were follows:1.The 18 parent materials had their own characteristics in plant height,tuber shape,skin color,flesh color and other traits;the dry matter content ranged from16.38% to 27.70%,and the starch content ranged from 8.62% to 23.19%,and the dry matter and starch content of P104 was the highest;protein content ranged from 0.87%to 2.92%;the reducing sugar content of the 18 parent materials were all below0.018%;the anthocyanin content ranged from 4.96 mg/kg·FW to 48.93 mg/kg·FW,and the anthocyanin content of P115 was the highest.2.The 10 pairs of SSR primers were used to amplify 18 parental materials,and obtained 118 loci,of which 103 were polymorphic loci,and the polymorphism ratio was 87.29%.The genetic distance(GD)ranged from 0.2329 to 0.6610,the average of GD was 0.4868.Based on the GD value of 0.52,the 18 parental materials were clustered into 3 categories: P57 was one category,P68,P114 and P121 were one category,P1,P7,P9,P12,P31,P79,P96,P99,P104,0112,P115,P117,P132 and P158 were one type.3.The nutritional quality of the 91 potato hybrids progeny were analyzed.The dry matter content ranged from 13.11% to 30.24%;the starch content ranged from9.97% to 27.56%;the protein content varied greatly,from 0.49% to 5.07%;The reducing sugar content were mostly ranged 0.008% to 0.027%,of which the sugar content of 33 hybrid plants were less than 0.018%;the anthocyanin content of 48 colored potato hybrids plants ranged from 1.32mg/kg·FW to 422.19mg/kg·FW,and the anthocyanin content of 2019084-37 was highest.4.Using two pairs of SSR primers STI027 and S153,the SSR banding patterns of 29 and 39 individual plants in the hybrid combinations 2019066 and 2019074 were divided into four types: parental complementation,deletion,paternal and maternal,and 28 and 37 true hybrids were identified respectively.The result indicated that SSR molecular marker technology was reliable for authenticity identification of potato hybrids. |
