The development of fine wool sheep’s skin and hair follicles directly affects the yield and quality.Therefore,studying on the regulation mechanism of fine wool wool’s follicles development is of great significance for improving wool yield and fineness,and it is also a very important content in the current breeding work of fine wool sheep.At present,epigenetic modification plays an important role in hair follicle development and it also can regulate gene expression.Transcription factors play an important role in the regulation of gene expression,and corresponding research is also one of the current hotspots.Therefore,epigenetic markers and their transcription factors that regulate the development in Subo Merino sheep and their genetic mechanisms can be studied from the epigenetic level.In this study,we used qRT-PCR to detect the expression levels of ITGB2,IGFBP4,and IGFBP2 genes in different fineness skin tissues of Subo Merino sheep;the DNA methylation modification status of ITGB2,IGFBP4,and IGFBP2 genes were detected using the BSP method,and the correlation analysis of the DNA methylation levels and expression levels of ITGB2,IGFBP4,and IGFBP2 genes was performed;the key transcription factors of ITGB2,IGFBP4,and IGFBP2 genes were predicted and screened by bioinformatics.Finally,the transcription factors screened by bioinformatics analysis,bioinformatics analysis was performed on the selected transcription factors in order to explore the regulation of target genes..The main results are as follows:(1)ITGB2,IGFBP4,IGFBP2 genes are expressed in different fineness skin tissues of Subo Merino sheep.Among them,the expression of ITGB2 and genes in extremely thick group skin tissues of Subo Merino sheep was significantly higher than that of extremely fine group skin tissues(P<0.01).The expression of IGFBP4 genes in extremely fine group skin tissues of Subo Merino sheep was significantly higher than that of extremely thick group skin tissues(P<0.01).The expression level of IGFBP2 gene in extremely fine group skin tissue of Subo Merino sheep was higher than that of extremely thick group skin tissue.(2)ITGB2 gene is highly methylated in the skin tissue of Subo Merino sheep,and the probability of methylation of extremely fine group skin tissue is higher than that of extremely thick group skin tissue;The IGFBP4 gene was hypomethylated in the skin tissues of Subo Merino sheep,and the methylation probability of extremely thick group skin tissue was higher than that of extremely fine group skin tissue;The IGFBP2 gene was hypomethylated in the skin tissue of Subo Merino shee.It was found that the DNA methylation of ITGB2 and IGFBP4 genes was significantly negatively correlated with their expression levels(P<0.01).(3)Based on bioinformatics analysis,6 key transcription factors JUN,EP300,SP1,PAX6,ETS1,and MYOD1 were screened out,laying a foundation for finding more transcription factors related to hair follicle development.In conclusion,this study analyzed the DNA methylation levels and expression levels of ITGB2,IGFBP4,and IGFBP2 genes in different fineness skin tissues of Subo Merino sheep and screened six key transcription factors JUN,EP300,SP1,PAX6,ETS1,MYOD1.This study provide theoretical basis for exploring epigenetic markers and transcription factors related to hair follicle development.
