| The Small Tail Han Sheep and Sunit sheep are one of the excellent local meat and fur sheep breeds in China,with fast growth and development,high adaptability and genetic stability of traits.However,the regulatory mechanisms affecting the carcass traits of Small Tail Han and Sunit sheep are still unclear,and the progress of selection and breeding by molecularly assisted markers is slow.Therefore,by exploring the candidate genes for the traits of thoracic vertebrae number and carcass length,which can significantly improve the carcass traits of livestock,we can accelerate the selection and breeding process of both Small Tail Han and Sunit sheep breeds,and thus improve the economic benefits of sheep industry.Based on some of the GWAS data obtained in the previous phase,important candidate genes that can significantly affect growth traits in sheep were obtained for multi-thoracic vertebrae traits and carcass length traits in Small Tail Han Sheep and Sunit sheep,namely OSBPL11,PAEP,ALDH1A2,KIAA2012,UTP20,HERC1,ALDH6A1,IQCA1,PARS2 and ANO7,and a total of 22 SNPs were identified.A total of 22 SNPs locus were identified.These locus were validated by SNP sequencing and other methods to expand the population,and correlation analysis was performed with related traits such as thoracic vertebrae number trait.In this study,383 Small Tail Han Sheep and Sunit Sheep with recorded rib number data,which could be divided into normal(T13)and mutant(T14)groups according to the number of thoracic vertebrae,were sampled from the longest dorsal muscle tissue,DNA was extracted,and fragment sequencing was performed for the said locus.The results showed that among these 10 candidate genes,OSBPL11,PAEP,ALDH6A1 and ALDH1A2,and IQCA1 each had 1 SNP with g.208040464A>G,g.3830771G>A,g.89180657T>C,g.53608636G>A,and g.4692837G>A,respectively.KIAA2012 has three SNPs,g.218577177A>G,g.218560972G>A,g.218577024A>T.UTP20 has three SNPs,g.182904872A>G,g.182909238G>A,g.182923455G>A.HERC1 has three SNPs,g.47373236G>A,g.47394074T>G,g.47398452A>G.PARS2 has four SNPs,g.30789785C>T,g.30789946T>A,g.30789995C>T,g.30790544T>C.ANO7 has four SNPs,g.535119G>A,g.535343T>C,g.541221C>T,and g.541186T>C,respectively.Among them,the locus found to be significantly correlated(P<0.05)with carcass length in Small Tail Han Sheep were KIAA2012 g.218577177A>G and HERC1 g.47373236G>A.In Sunit sheep,the locus significantly associated with the number of thoracic vertebrae(P<0.05)were ALDH1A2 g.53608636G>A and UTP20 g.182909238G>A.The remaining 18 SNPs were not significantly associated with the number of thoracic vertebrae and carcass length traits(P>0.05).In conclusion,the four SNPs locus KIAA2012 g.218577177A>G,HERC1 g.47373236G>A,ALDH1A2 g.53608636G>A and UTP20 g.182909238G>A can be used as auxiliary selection markers potentially affecting sheep carcass traits for improving sheep production performance and preparing screening The four SNPs can be used as potential auxiliary selection markers for sheep carcass traits,and provide data support for improving sheep performance and preparing screening kits for traits specific to sheep thoracic vertebrae and carcass length. |
