| Temperature is one of the important factors affecting the normal growth and development of peanut.The growth period of spring peanut may encounter many kinds of extreme weather,such as cold in late spring,so as to prolong the germination time of peanut seeds and the growth of seedlings,which may seriously cause some seeds to lose their germination ability.Therefore,it is of great significance to take effective measures to promote the normal germination of peanut seeds at low temperature.In this experiment,"Baisha 1016"and"yuanza 9102"were used as materials,and cultured in an artificial climate chamber.By measuring the changes of morphological indexes,membrane lipid peroxidation(MDA),antioxidant scavenging activity and non enzymatic antioxidant system,the Alleviative Effect of hydrogen peroxide(H2O2)pretreatment on the low temperature inhibition of peanut seed germination was explored.The test results are as follows:(1)The germination of peanut seeds was inhibited obviously under low temperature. Through the study of the vigor indexes of peanut seeds,such as the dew rate and germination rate,it can be seen that the low temperature has a more obvious inhibitory effect on peanut seeds.The results showed that the germination rate of Baisha 1016 was inhibited by 42.82%and yuanza 9102 by 23.82%.(2)Pretreatment with hydrogen peroxide(H2O2)can obviously alleviate the damage of low temperature to peanut seeds.The results showed that the germination rate of peanut germplasm Baisha 1016 increased by 20%and yuanza 9102 by 22.54%after soaking in hydrogen peroxide.The results showed that the pretreatment with 1%H2O2could alleviate the morphological index of peanut seed germination inhibited by low temperature.seven hundred and forty-one million two hundred and five thousand three hundred and sixty-two(3)The results showed that exogenous concentration of 1%H2O2could significantly inhibit the accumulation of MDA in peanut seeds.The content of MDA in plant decreased and the damage degree of cell membrane structure decreased.Hydrogen peroxide(H2O2),as a kind of reactive oxygen species(ROS),accumulates in plants under low temperature.It was found that the MDA and H2O2contents of the two peanut seeds increased significantly with the delay of treatment time.In Baisha 1016,the content of MDA and H2O2decreased by 74.12%and 73.1%,respectively.After yuanza 9102 was treated,the content of MDA and H2O2decreased by 56.32%and 40.4%respectively.The results showed that the MDA content and H2O2Content of peanut seeds pretreated with 1%H2O2were significantly lower than those of untreated seeds.(4)In low temperature treatment,exogenous hydrogen peroxide(H2O2)of certain concentration can effectively improve the activity of antioxidant enzymes in peanut seeds,and achieve the role of scavenging ROS accumulation.The pretreatment with H2O2can significantly inhibit the increase of H2O2Content,and the application concentration of 1%H2O2can also significantly improve the activities of superoxide dismutase(SOD),catalase(CAT),peroxidase(POD)and ascorbic acid peroxidase(APX)in peanut seeds,fully remove MDA,H2O2and other toxic substances in seeds,and maintain ROS The metabolism balance can reduce the negative effect of lipid peroxidation in the cell membrane of seeds,so as to enhance the low temperature resistance of seeds. |
