In this study,a total of 21 populations of perennial tritelytrigia germplasm lines were used as research materials,which was expanded by means of root division propagation and direct seeding breeding.The cold resistance,perennial characteristics and genome composition of perennial tritelytrigia germplasm lines and propagation populations were analyzed and identified by means of morphological analysis and molecular cytogenetics.The purpose of this study is to clarify the chromosomal composition and genetic characteristics of different types of perennial tritelytrigia,so as to provide material basis and theoretical basis for the study of cold resistance and perennial characteristics of perennial tritelytrigia in cold areas.The results of morphological investigation showed that 21 perennial tritelytrigia were perennial,cold resistant and regenerated after cutting.The plants grew luxuriantly and the morphological characters tended to be stable.The plant height was more than 1m,and the number of tillers was increased and up to 648.The seed setting rate was generally stable except 19-51820443.The seed setting rate of 19-425299128was the highest and more than 88%.There were six materials that the 1000 grain weight was more than 6g,and the highest of 1000 grain weight was 19-412138 with8.86g.Moreover,a total of 52 expanded materials were obtained,and their morphological characteristics were consistent with those of the original plants.The results of cytology and molecular markers showed that the chromosome number of somatic cell from the root tips was more than 47 in 19-42529356,and the chromosome number of 19-51820443 was 42-44.The chromosome numbers of somatic cells from other materials were 42.The chromosome numbers of expanded materials were the same as those of original plants,and they also had genetic components of Thinopyrum intermedium with molecular marker test.However,non-specific bands were amplified from some perennial tritelytrigia and propagation materials,indicating that there were differences in genetic components among materials.The results of GISH analysis showed that the genome composition of most plants was St St JJJvsJvsthat was similar to Th.intermedium,but thehybridization signals on chromosomes regions were slightly different.Among the identified materials,19-42529356 and 19-51820443 carried 11 and 1 common wheat chromosome,respectively.This result provided a new direction for the study of perennial tritelytrigia germplasms in the future.
