Objective To investigate the effects of human amniotic mesenchymal stem cells(h AMSCs)transplantation on expression of inducible nitric oxide synthase(i NOS)after rat with Spinal cord injury(SCI).Methods Thirty six female adult SD rats weighing 250-300 g were randomly divided into three experimental groups,each group containing 12 experimental bodies.Three groups were named sham group(sham group),control group(SCI group)and cell transplantation group(hmascs group).In sham group,only laminectomy was performed to expose the spinal cord without injury,at the same time acute spinal cord injury model was established in SCI group and hmamscs group by modified Allen’s method.The rats of SCI group were injected with hosphate buffered saline(PBS),and the rats of h AMSCs group were injected with h AMSCs.On the 4th day after operation,the tail vein blood of rats in each group was collected,and the spinal cord tissue samples were collected.The expression of i NOS in the spinal cord was detected by Western Blot and immunohistochemistry.Determination of the concentration of NO in blood samples by nitrate reductase method.Western blot and IHC showed that compared with sham group,i NOS protein expression in SCI group and h AMSCs group was significantly higher(P<0.05),and i NOS protein expression in h AMSCs group was lower than that in SCI group(P<0.05).IHC results showed that immunopositive staining was located in the cytoplasm of neurons and astrocytes,and was widely distributed in the front foot,back foot and around the central canal of spinal cord gray matter;there were also a few positive staining in the white matter of spinal cord.Among the three groups,the blood NO content in sham group was the lowest,while that in SCI group and h AMSCs group was higher(P<0.05).NO content in blood of SCI group was higher than that of h AMSCs group(P<0.05).ResultsConclusions Transplantation of human amniotic mesenchymal stem cells after acute spinal cord injury in rats can reduce the expression of inducible nitric oxide synthetase,and then reduce the production of nitric oxide and its content in blood,which may have an effect on the secondary inflammatory pathological process of nerve tissue after acute spinal cord injury in rats.
