Objective:To explore the dependency between soluble low density lipoprotein receptor 11(sLR11)gene single nucleotide polymorphism(SNP)of the rs689021 site and sLR11 levers with type 2 diabetes mellitus(T2DM)and carotid atherosclerosis in Korean nationality area of Yanbian,China.Subjects and methods:All subjects were from 313 patients admitted to the hospital of Yanbian university in Jilin Province from September 2019 to January 2021.According to the IMT value of carotid artery and the presence or absence of plaque,237 patients with T2 DM were summed up T2 DM with carotid atherosclerosis group(CAS group)according to carotid artery IMT ≥1mm.Patients with carotid artery IMT < 1.0mm but without plaque were divided into T2 DM without carotid atherosclerosis group(NCAS group,n = 104).In addition,Healthy subjects with normal glucose tolerance were selected as control group(NGT group,n = 76).The IMT value of the left and right side of the common carotid artery and the presence of atherosclerotic plaques in the common carotid artery were detected by color Doppler ultrasound.Genomic DNA was extracted from whole blood.Single nucleotide polymorphisms(SNPs)were typed and sequenced by PCR-LDR method.The level of sLR11 was detected by monoclonal antibody sLR11 kit.SPSS26.0 statistical software was used for data analysis.Results:1.NGT group,NCAS group and CAS group were all C < T,and genotype frequencies were all CC < TT < CT,At the same time,there was no significant difference in rs689021 genotype frequency and allele of sLR11 gene among NGT group,NCAS group and CAS group(P>0.05).2.The statistical results of rs689021 genotypes CC,CT and TT showed that the body weight and BMI in NCAS group in TT and CC group were lower than those in CT group,and SBP in TT group was lower than that in CC group in NCAS group(P<0.05).3.The levels of BMI,SBP,FPG,TG and sLR11 in CAS group and NCAS group were obviously higher than those in NGT group,DBP in NCAS group was higher than that in NGT group,and TC and LDL-C in CAS group were obviously higher than those in NGT group(P<0.05).4.Gender(male),smoking,FPG,TG,HDL-C and LDL-C correlated with IMT thickening.Among them,OR < 1 of HDL-C was the protective factor of IMT thickening,and the OR values of LDL-C,FPG and were all > 1,which were risk factors of IMT thickening.There was statistical difference(P < 0.05).5.Age,SBP,FPG,TG,sLR11,IMT ≥ 1mm,renal insufficiency,coronary heart disease,cerebrovascular disease and atherosclerotic plaque were correlated.The OR values of age,FPG and IMT ≥1mm were all > 1.These factors are the risk factors of atherosclerotic plaque.There was statistical difference(P < 0.05).6.Gender(male),smoking,age,BMI,SBP,FPG,TC,TG,atherosclerotic plaque and sLR11 level were correlated.Among them,the OR values of age,BMI,FPG and TC were all greater than 1,which indicated that these factors were the risk factors of sLR11 level.There was statistical difference(P < 0.05).Conclusion:1.The rs689021 polymorphism of sLR11 gene was not related to the occurrence of T2 DM.2.The rs689021 polymorphism of sLR11 gene is not related to the occurrence of T2 DM complicated with CAS.3.The level of sLR11 is correlated with age,BMI,FPG and TC.SLR11 may increase the risk of carotid atherosclerosis in T2 DM by up-regulating the levels of blood glucose and blood lipids.
