Objective:To investigate the correlation between sLR11(Soluble low- density lipoprotein recptor fami- ly memberll) single nucleotide polymorphism and atherosclerosis in patients with type 2 diabetes mellitus in two ethnic groups (Korean nationality and Han nationality) in Yanbian region.Research Object:During the period from December 2012 to December 2015 in Yanbian University Affiliated Hospital,350 cases with type 2 diabetes mellitus were selected from the outpatient and inpatient departments of Endocrine Department of Internal Medicine. According to the carotid IMT values the cases were divided into (1) carotid atherosclerosis group (CAL group, n= 163) in which carotid IMT> 1.0 mm of plague (2) non-carotid atherosclerosis group (NCAL, n = 187) where carotid IMT< 1.0 mm, (3) normal control group (healthy) (NGT, n=172).Methods:For the measurement of carotid IMT and atherosclerosis plaque by using color ultrasonic diagnostic apparatus. SNP typing and sequencing are done by LDR-PCR Technology. Using Hardy-Weinberg’s law of equilibrium, the group representative of the study sample is determined. One-way ANOVA was used to compare the measurement data between the two groups where pair wise comparisons were done by LSD method. Logistic regression analysis was used to screen the risk factors of carotid atherosclerosis, with P< 0.05 as a statistical significance.Results:1. The frequencies of allelic genes A and T were 54.3% & 45.7%,56.7% & 43.3%,58.1% & 41.9%, respectively, in the CAL group, the NCAL group and the NGT group in the Yanbian region. In CAL, NCAL and NGT groups, AA genotype frequency was 27.6%,19.0% and 30.2%, the AT genotype frequency was 53.4%, 32.1% and 55.8%, the TT genotype frequency was 19.0%,32.1% and 14.0%. The CAL group and NGT group, NCAL group and NGT group, CAL group and NCAL group had no significant difference in genotype and allele frequency distribution (P>0.05). There were no significant differences between the groups dominant pattern and recessive pattern of each genotype and allele frequencies(P>0.05).2. In NGT, NCAL and CAL groups, in Korean nationality patients, frequencies of allelic gene A was 53.8%,53.3% and 50.7%, T was 46.2%,46.7% and 55.3%, AA was 26.2%,23.7% and 24.3%, AT was 55.4%,59.2% and 52.7%, the TT was 18.5%,17.1% and 23.0% where as in Han nationality frequencies of allelic gene A was 60.5%,53.3% and 55.4%, T was 39.5%,46.7% and 44.7%, AA was 33.0%, 36.8% and 27.1%, AT 55.0%,43.4% and 56.5%,TT 12.0%,19.8% and 16.5%. Statistically, there was no significant difference between all three groups of Korean and Han population genotype and allele frequency (P>0.05).3. The regression coefficients SBP, LDL-C and HbAlc were respectively 0.024, 0.585, and 0.251 which shows SBP, LDL-C, HbAlc were the risk factor for carotid atherosclerosis. It is positive correlation between regression coefficient and carotid atherosclerosis.4. Three genotype group in type 2 diabetes with carotid atherosclerosis were performed statistical analysis,including age and metabolic parameters.The level of CHO is highest in AT genotypes group than AA and TT genotype group, significant difference(P<0.05). The level of CHO is higher in AT genotypes group than TT genotype group, significant difference(P< 0.05).The level of TG in AT genotype group is higher than in TT genotype group, statistically significant (P< 0.01).The level of LDL-C is highest in TT genotype than in AT and AA genotype group, significant difference(P<0.01). The level of LDL-C is higher in TT genotype than in AA genotype,statistically significant (P< 0.01).Conclusion:1. There was no significant correlation between sLRll gene rs3824968 polymorphism and type 2 diabetes mellitus in Yanbian region.2.sLR11 gene rs3824968 polymorphism among Korean and han has no racial differences.3.sLRll gene rs3824968 polymorphism T factor may affecte the degradation of LDL, consequently increase the risk of T2DM with carotid atherosclerosis.4. Systolic blood pressure, low density lipoprotein cholesterol, glycosylated hemoglobin are the risk factors for carotid atherosclerosis.
