Study On The Correlation Between CT Texture Characteristics Of Lung With Abnormal Liver Perfusion And Reduced Intestinal ICC In Paraquat Poisoning Rabbits

Objective:To investigate the correlation analysis of lung CT texture characteristics with abnormal changes of liver perfusion imaging parameters(artery flow(AF),portal flow(PF),perfusion index(PI))and the degree of small intestinal Cajal interstitial cell(ICC)decrease after rabbit paraquat poisoning induced multiple organ function injury.Methods:Twenty-four healthy adult rabbits were randomly divided into control group(0.9%NaCl)and paraquat(35mg/kg)group.2h,24h,7d and 14d after administration,320-row lung volume CT plain scan and liver perfusion imaging scan were performed in each group.CT texture characteristic parameters of both lungs of experimental rabbits in each group were extracted at each time point,and the values of liver perfusion imaging parameters(AF,PF and PI)were measured on the pseudo color CT liver perfusion images,respectively.At the same time,2 rabbits in each group were randomly killed.Lung and liver tissues were dissected and extracted for routine pathological examination,and the percentage of ICC in the small intestinal wall was obtained by immunohistochemistry.The correlation between lung CT texture features,liver perfusion imaging parameters and ICC values of small intestine was analyzed by Pearson correlation,and the quantitative relationship was obtained.Results:1.Pathological changes of lung,liver and small intestine:1)Under light microscope,the tissue structure of bronchus and alveoli in both lungs of control group was clear,and no inflammatory cell infiltration was observed.The pathological changes of lung tissue in the PQ group at 2h to 14d were as follows:a small amount of epithelial cells could be seen in the local bronchial lumen of the lung,and more lymphocytes could be seen around it.Hemorrhage was seen locally in the lung,and more red blood cells were seen in alveolar lumen and bronchial lumen.There were a few goblet cells and more lymphocyte infiltration around the bronchial epithelium in the local lung.The result is severe local bleeding in the lungs and a large number of red blood cells in the alveolar space.2)Under light microscope,the structure of hepatic lobules was clear,the hepatic cords were arranged neatly,and the hepatic sinuses were not dilated or squeezed.The pathological changes of liver tissue in PQ group from 2h to 14d were as follows:the number of white blood cells in hepatic sinuses increased;Hepatic sinus congestion,more red blood cells were seen in hepatic sinus;Pontate infiltration of lymphocytes around bile duct in local portal area;Finally,hepatic sinus congestion and dilatation,the local tissue margin is more serious.3)The results of immunohistochemistry showed that the percentage of intestinal ICC in PQ group was significantly decreased with time prolonging.2.Imaging changes of lung CT:The bronchial vascular bundles of experimental rabbits in both lungs were clear at each time point in the control group,and no obvious exudation or consolidation images were observed.In the paraquat group from 2h to 14d,the plain CT imaging changes of the experimental rabbits’ double lungs showed that at the early stage,the lung texture was thickened and blurred,and a little ground glass shadow was changed,etc.With the extension of time,local lung consolidation shadow was observed as the lesions became increasingly aggravated.In the late stage,the main changes were pulmonary fibrosis,and serious complications such as pneumothorax could be seen.3.Changes of lung texture analysis parameters:Compared with the control group,the lung CT texture analysis parameters of paraquat group showed a decreasing trend with time.The mean,kurtosis,skewness,percentile(Perc.1%,Perc.10%,Perc.50%,Perc.90%,Perc.99%),short run,long run unevenness and stroke ratio increased with time.4.Changes in liver perfusion parameters:Compared with the control group,the values of AF,PF and PI of liver perfusion parameters in the paraquat group showed a decreasing trend over time.5.Correlation analysis of lung CT texture analysis parameters with changes in liver perfusion parameters and percentage of intestinal ICC:1)Compared with the control group,there was a linear correlation between liver perfusion parameters(AF,PF,PI)and four lung CT texture analysis parameters in Paraquat group,and AF was positively correlated with mean value and Perc.50%(r=0.4751,0.5288,P<0.05).And negatively correlated with kurtosis and skewness(r=-0.5899,-0.6927,P<0.05).Pf was negatively correlated with kurtosis and skewness(r=-0.5357,-0.6516,P<0.05).Pi was negatively correlated with kurtosis and skewness(r=-0.5393,-0.6371,P<0.05).2)Compared with the control group,the percentage change of intestinal ICC in paraquat group was linearly correlated with liver perfusion parameters(AF,PF,PI)and six lung CT texture analysis parameters.The percentage of ICC in small intestine was positively correlated with AF,PF,PI,mean value and percentile(Perc.1%,Perc.10%,Perc.50%)(r=0.8110,0.7766,0.7361,0.5204,0.5183,0.5591,P<0.05).Was negatively correlated with kurtosis and skewness(r=-0.6339,-0.6871,P<0.05).Conclusions:1.Lung CT texture characteristic parameters,liver perfusion parameters and intestinal ICC percentage values can be used to quantitatively evaluate lung injury,liver perfusion and intestinal injury after PQ poisoning;2.There is a certain correlation between lung injury and liver perfusion and intestinal injury after PQ poisoning,and the degree of injury of liver perfusion abnormality and intestinal function can be understood through the changes of texture characteristics of lung CT plain scan.
