Establishment of an appropriate model of paraquat provide the necessary conditions for the follow-up study.70healthy New Zealand white rabbits were randomly divided into four groups,10for group, exposed to different doses of paraquat, respectively,25mg paraquat for the first group,35mg for the second group,45mg for the third group, the fourth group does is50mg, which is the median dose lethal. Preset time1h,6h,12h,24h,48h and72h, respectively, selected three rabbits in each group and took six lungs as six specimens,(1)HEstaining were detected by light microscopy.(2) ultrastructure was observed by microscopy, paraquat35mg.kg is the optimal dose, as the follow-up CT perfusion stable and reliable model of chemical-induced lung injury. Study early changing of hemodynamics in acute chemical lung injury through320-slice CT volume perfusion, and compare with the pathological Correlation, then seek the value of early monitor and diagnosis in CTP early functional change of chemical pulmonary injury.30New Zealand white rabbits were divided into control group and experimental group.(control group is0hour)The rabbits in experimental group were treated with PQ at35mg/kg through intraperitoneal injection. Experimental groups were scaned chest by320-slice CT(0,2,6,12,24,48h)at the corresponding time and0hour rabbit were killed by air embolism(n=4) and obtained the lung tissues for pathological exam.(1) CTP average parameter, rBF, rBV, rPS of the control group were128.54±31.31、25.20±4.15、13.06±3.05;(2)there was a significant correlation between the changes of experimental group CTP parameter and pathohistology. The rank of capillary endothelial cellular swelling has a negative correlation with the volume of the blood flow, r=-0.848, p=0.033. The rank of capillary basement membrance destorying has positive correlation with vasopermeability, r=0.893, p=0.016.After injecting paraquat, The pulmonary blood flow decreases gradually by time. Analysis of variance predicts that volume of blood flow decreases significantly in contrast with the blood flow when no injecting in24hours, p<0.001. After injecting paraquat, the permeability of pneumoangiogram upgrades gradually by time. Analysis of variance predicts that there is significant variation in the permeability of pneumoangiogram in contrast with it when no injecting paraquat in2hours, p<0.01. CTP parameters (especially rPS) may reflect the hemodynamic change of paraquat induced chemical pulmonary injuryin the early stage, It is valuable for in predicting chemical pulmonary injury in early time.
