| Objectives:The incidence of obesity is high globally,but the concern about obesity among ethnic minorities in China is low,especially in Yunnan.FA components can regulate appetite and energy metabolism,weight gain may be closely related to the body’s plasma high FA concentration caused by lipid metabolism disorders,it is still necessary to verify whether plasma FA is associated with increased dietary FA intake.By establishing an optimized GC/MS method to detect plasma FA,this study analyzes and compares the plasma FA composition of four unique ethnic minority obese populations in Yunnan,further analyzes the relationship between diet FA and plasma FA in obese patients,and its association with obesity,with the aim of providing reference indicators and theoretical basis for the clinical diagnosis and treatment of obesity.Methods:The study relied on the NSFC’s "Study on the Relationship between Four Unique National FA,Visfatin,A-FABP,and PPAR X2 Genes pro12Ala,C161→T Polymorphism and Chronic Diseases in Yunnan"Baseline survey results,from 4 unique ethnic minorities 1913 people screened obesity survey subjects(BMI≥28.0kg/m~2)as the obesity group,according to ethnic consistency,gender consistency,age(±2 years old),match the normal group of respondents(18.5kg/m~2≤BMI<24.0kg/m~2)by 1:1,using case-control studies to collect basic Information,obtaining health check-up data through medical examination and laboratory testing,obtaining dietary intake of residents through food frequency questionnaires,calculating dietary FA intake using Excel 2016 editing formula,and using optimization method GC/MS technology to detect plasma FA concentration Using Epidata3.0 software to establish database input data,orthogonal design assistant Ⅱ to design and analyze orthogonal test results,Using IBM SPSS26.0 statistical software for statistical analysis,P<0.05 as the difference is statistically significant,and GraphPad prism6 software drawing.Results:1.Among the three factors affecting the temperature,sulphuric acid concentration and time of plasma FA methylesteration,the influencing factors ranged from large to small:temperature>sulphuric acid concentration>time,in which the temperature has statistical significance to the difference of plasma FA derivative effect,determine 7%sulphuric acid concentration The correlation coefficient range of the standard curve of fatty acids ranged from 0.9991 to 0.9998 and the detection limit range was 0.01 to 0.41 mg/L.The intraday CV value is 0.27 to 6.15%,the daytime CV value is 0.91 to 8.70%,and the intraday and daytime CV value is less than 10.0%.The recovery rate was 80.06%to 118.34%,both between 80.00%and 120.00%,indicating that the stability and accuracy of the method were good.2.In the Hani population,plasma C16:0 and C18:2 n-6c in the obesity group were higher than in the normal group(P<0.05).In the Naxi population,the normal plasma C18:0 and C20:0 content was higher than that of the obesity group(P<0.05),while the plasma C14:0,C17:0,C18:2 n-6c,C11 in the obesity group 8:3 n-6,C18:3 n-3,C20:3 n-6,C20:4 n-6,C20:5 n-3 and total FA higher than normal group(P<0.05).In the Bulang population,the obese group had plasma C16:0,C16:1 n-7,C18:1 n-9c,C20:3 n-6,C20:5 n-3,and the total FA was higher than normal(P<0.05).In the Wa population,the plasma C14:0,C16:0,C16:1 n-7,C18:1 n-9c,C20:3 n-6,C20:5 n-3 were higher than the normal group(P<0.05).Comparing plasma FA distribution differences among 4 ethnic minorities,of which the total FA level of Naxi plasma was higher than that of Hani(P<0.05),and in the analysis of individual FA,two comparisons found that 12 plasma FA had the highest content in the Naxi ethnic group,and 11 plasma FA had the lowest content in Hani.3.Naxi(typical correlation coefficient:0.676),Bulang(typical correlation coefficient:0.879 to 0.974)and Yunnan 4 unique ethnic minorities(typical correlation coefficient:0.0 473-0.787)There is a close positive correlation between plasma FA and dietary FA(P<0.05),which means that the higher the dietary FA level ingested in the diet,the higher the plasma FA level.There was no correlation between plasma FA and dietary FA in Hani and Wa populations(P>0.05).4.Plasma C16:0(OR=1.310,95%CI:1.028~1.669)may be a risk factor for obesity in the Hani population,i.e.those with higher plasma C16:0 levels are 1.310 times more likely to be obese than those with lower plasma C16:0 levels.Diastolic pressure(OR=1.054,95%CI:1.006~1.103)may be a risk factor for obesity in the Naxi population,i.e.the likelihood of obesity occurring in people with higher diastolic pressure levels is diastolic pressure levels The lower 1.054 times;plasma C18:0(OR=0.499,95%CI:0.343~0.727)may be a protective factor for obesity in the Naxi population.Plasma C20:3 n-6(OR=6.250,95%CI:1.224~31.927)and diet C20:1(OR=8.879,95%CI:1.126~69.989)may be a risk factor for obesity in the Wa population,i.e.people with high plasma C20:3 n-6 and dietary C20:1 levels had 6.250 and 8.879 times more on obesity than those with low levels.Systolic pressure(OR=1.045,95%CI:1.022~1.068),plasma C20:3 n-6(OR=2.523,95%CI:1.350~4.716),plasma C20:5 n-3(OR=3.614,95%CI:1.412~9.251)may be a risk factor for obesity in the study population as a whole,i.e.Those with higher levels of systolic blood pressure,plasma C20:3 n-6 and plasma C20:5 n-3 were 1.045,2.523,3.614 times more likely to be obese than those with lower levels;and HDL(OR=0.275,95%CI:0.087~0.871)may be a protective factor for obesity in the study population as a whole.Conclusions:1.The temperature,sulfuric acid concentration and time during the pretreatment of plasma samples will affect plasma fatty acid derivation,the temperature effect is large,GC/MS is a favorable tool for detecting plasma FA,with good separation,accuracy and repeatability.2.The fat group of Hani,Naxi,Bulang and Wa had higher plasma FA than normal groups,and there were differences in the distribution of plasma FA among the four ethnic minorities,of which the total fatty acid level of Naxi plasma was the highest and the total fatty acid level of Hani plasma was the lowest.3.There is a positive correlation between plasma FA and diet FA among 4 unique ethnic minority populations in Yunnan,and the higher the dietary FA level,the higher the plasma FA level,but there are ethnic differences,the Naxi and Bulang ethnic groups are closely related,Hani and Wa are not related.4.There are differences between plasma FA and fat fat in 4 unique ethnic minority populations in Yunnan,plasma palmic acid(C16:0)in Hani population may be a risk factor for obesity,plasma stearic acid(C18:0)in Naxi population may be a protective factor for obesity,and plasma DGLA(C20:3 n-6)in Wa population may be a risk factor for obesity. |
