Objective:To observe the effects of theanine on(HRCECs)proliferation and vascular endothelial growth factor(VEGF)expression of human retinal vascular endothelial cells in high glucose environment.Method:human retinal vascular endothelial cells were cultured in vitro and divided into 6 groups: group D(low glucose control group),5.5 mmol glucose control group(high glucose control group),25 mmol glucose control group(L1group),L2 group(H group + 50 μmol/L theanine),L3 group(H group + 100μmol/L theanine)and L4 group(H group + 200 μmol/L theanine).HRCECs was treated with theanine at different concentrations mentioned above.After 24 hours,48 hours and 72 hours,the proliferation of HRCECs was observed by MTT method,and the expression of VEGF in HRCECs was detected by Western blotting(Western-blot,WB).Result:1.The results of MTT assay showed that: 1)Under the same culture time,compared with group D,the cell proliferation activity of group H was primarily higher than that of group D,P < 0.05,which was statistically significant.2)in the same culture time,after the intervention of different concentrations of theanine,with the increase of theanine concentration(concentration gradient of 25 μmol/L,50 μmol/L,100 μmol/L,200 μmol/L,respectively),L1 group,L2 group,L3 group,L4 group were compared with H group,respectively.Within a certain concentration range,the inhibition rate of HRCECs increased with the increase of theanine concentration(P<0.05).3)when the cells were treated with the same concentration of theanine at different time points,the inhibition rate of HRCECs increased with the extension of time in a certain time range,P<0.05,which was statistically significant.2.WB method showed that: 1)under the same culture time,compared with group D,the expression of VEGF in group H wasprimarily higher than that in group D(P<0.05).2)under the same culture time,after the intervention of different concentrations of theanine,with the increase of theanine concentration(concentration gradient of 25μmol/L,50 μmol/L,100 μmol/L,200 μmol/L,respectively),L1 group,L2 group,L3 group,L4 group were compared with H group,respectively.Within a certain concentration range,the expression of VEGF in HRCECs decreased with the increase of theanine concentration(P<0.05).3)when the cells were treated with the same concentration of theanine at different time points,the expression of VEGF in HRCECs decreased with the extension of culture time in a certain time range,P<0.05,which was statistically significant.Conclusion:1.Theanine can inhibit the proliferation of human retinal vascular endothelial cells in a high glucose environment in vitro,with a certain concentration and time dependence;2.Theanine can down-regulate the expression of VEGF in human retinal vascular endothelial cells under a high glucose environment in vitro,Has a certain concentration and time dependence.
