Effects Of Circ_0001809 On Proliferation,migration,invasion And Apoptosis Of Human Osteosarcoma Cells | | Posted on:2022-04-29 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y C Zou | Full Text:PDF | | GTID:2494306344989699 | Subject:Clinical Medicine | | Abstract/Summary: | PDF Full Text Request | | Objective:By studying the effects of circ_0001809 on the proliferation,migration,invasion and apoptosis of osteosarcoma cells,the role circ_0001809 in the progression of osteosarcoma was clarified.Method:Clinical samples(tumor tissues and adjacent tissues)of oste osarcoma patients,osteosarcoma cell line MG-63,Saos-2,and the human osteoblast cell line h FOB1.19 were used as research objects.The expression level of circ_0001809 in clinical samples was measured by q RT-PCR.The expression level of circ_0001809 in cells was detected by PCR;si-circ_0001809 and p CMV-circ_0001809 were transfected into MG-63 and Saos-2 cells,respectively.Proliferation,migration,invasion and apoptosis of MG-63 and Saos-2 cells were detected by CCK-8 assay,scratch assay,Transwell assay and flow cytometry.Results:1.The expression level of circ_0001809 in clinical samples of osteosarcoma tissues was significantly higher than that in adjacent tissues(P<0.001).2.The expression level of circ_0001809 in MG-63 and Saos-2 cells was significantly higher than that of h FOB1.19 cells(P < 0.05).3.CCK-8 results showed that the low expression of circ_0001809significantly inhibited the proliferation of MG-63 and Saos-2 cells compared with the control group cells(P<0.001);in contrast,high expression of circ_0001809 promoted the proliferation of MG-63 and Saos-2 cells(P<0.001).4.The scratch test showed that,compared with the control group cells,low expression of circ_0001809 inhibited the migration of MG-63 and Saos-2cells(P<0.001);in contrast,high expression of circ_0001809 promoted the migration of MG-63 and Saos-2 cells(P<0.001).5.Transwell assay results showed that,compared with the control group,the low expression of circ_0001809 significantly inhibited the migration(P<0.001)and invasion(P<0.001)of MG-63 and Saos-2 cells;high expression of circ_0001809 promoted tthe migration(P<0.05)and invasion(P<0.001)of MG-63 and Saos-2 cells.6.The results of flow cytometry showed that,compared with the control group,the apoptosis rate of the circ_0001809 low-expression group was increased(P<0.05)the apoptosis rate of the circ_0001809 high-expression group was decreased(P<0.05).Conclusion:1.Circ_0001809 was differentially and highly expressed in human osteosarcoma tissues;2.Circ_0001809 promoted the proliferation,migration and invasion of MG-63 and Saos-2 cells,and inhibited cell apoptosis. | | Keywords/Search Tags: | circ_0001809, osteosarcoma, proliferation, migration and invasion, apoptosis | PDF Full Text Request | Related items |
