Effects Of YAP On Proliferation,Apoptosis,Invasion And Migration Of Osteosarcoma Cells

Objective : Osteosarcoma is a primary malignant bone tumor,which generally originates from bone tissue cells.Osteosarcoma is a kind of malignant tumor with a high incidence,especially in children and adolescents,which seriously damages physical and mental health.Its pathophysiological changes are as follows: tumor cells proliferate to form bone like tissue or immature bone.Its incidence rate accounts for about 20% of primary bone malignant tumors,and the metastasis is early,the degree of malignancy is high,with a high disability,fatality rate,has attracted extensive attention at home and abroad.Osteosarcomas are prone to occur in areas with abundant bone blood supply,such as the metaphysis of the long shaft,and occasionally in other areas.The incidence of hematogenous metastasis is high,the metastasis is early,and the progression is rapid.With the method of treatment of osteosarcoma in recent years has made great progress,especially the rapid development of chemotherapy significantly reduces the outside the carcinoma metastasis rate,its total 5 years survival rate also greatly improve,to improve the quality of life of patients with a certain,but the current clinical use methotrexate,more gentle than star,cisplatin and other drugs to treat osteosarcoma,after the chemotherapy drug in some patients prognosis is still poor,appear even drug resistance,even if combined with other treatments such as surgery and radiotherapy is difficult to improve prognosis and quality of life.On the other hand,in the use of large doses of chemotherapy drugs at the same time,also produced serious toxic side effects,resulting in double harm,to bring great physical and mental pain to patients.At present,gene therapy is one of the hot spots in current tumor therapy,which is expected to cure and prevent tumors at the root.Therefore,in-depth study on the gene and molecular mechanism of the occurrence and development of osteosarcoma and search for effective targets of osteosarcoma gene and molecular therapy will provide new methods for the effective treatment of osteosarcoma.To investigate the Effects of YAP on proliferation,apoptosis,invasion and migration of osteosarcoma cells.Methods : Human osteosarcoma cell lines U2 OS and MG63 were cultured in a modified DMEM/ high-glucose medium containing 10% fetal bovine serum.We constracted the recombinant plasmid of sh YAP and pc DNA Flag YAP.Then the empty plasmid and the recombinant plasmids were transfected into human osteosarcoma cell lines,the expression of YAP in MG63 and U2 OS was detected by Western blot,and cell proliferation,migration,invasion and apoptosis were detected by using methyl thiazol tetrazolium(MTT)assay,Transwell migration assay,Transwell invasion assay and flow cytometry assay,respectively.Results:The abilities of proliferation,invasion and migration of U20 S and MG63 cells transfected with pc DNA Flag YAP were significantly increased,and the cells apoptosis was significantly reduced,The abilities of proliferation,invasion and migration of U20 S and MG63 cells transfected with sh YAP were significantly reduced,and the cells apoptosis was significantly increased(P<0.05).Conclusion:The overexpression of YAP can promote the proliferation,invasion and migration,and reduced apoptosis of osteosarcoma cells,YAP somehow enhanced the proliferation,migration and invasion ability of osteosarcoma cells U2 OS and MG63,and weakened their apoptosis.This study indicated that YAP could be used as a potential target for early diagnosis and treatment of osteosarcoma,laying a foundation for further study on the role of YAP in the pathogenesis of osteosarcoma.
