Establishment Of Choledochojejunostomy Model In Young Rabbits And The Study Of Postoperative Gastrointestinal Function

Objective: To study the changes of gastrointestinal motility,the number of interstitial cells of Cajal(ICC)and the gene and protein expression of related regulatory factors in young rabbits after choledochojejunostomy.Methods: Ninety 1-month-old rabbits were divided into three groups: control group,long jejunal branch group and short jejunal branch group,with 30 rabbits in each group.The rats were sacrificed after enema at different time points of 1 day,7 days,14 days,1 month,3 months and 6 months after operation.The number of ICC cells was identified by immunofluorescence.The level of mscf was detected by Western bolt.The expression of ckit in ICC was detected by laser scanning confocal microscope.Reverse transcription polymerase chain reaction(RT-PCR)was used to detect the m RNA expression of ckit,mscf and TMEM16 A of Ano1.Results: before the operation,there was no significant difference in the average body weight of each group(P > 0.05).From 1 day to 6 months after the operation,the body weight,gastric emptying rate and intestinal propulsion rate of the young rabbits increased gradually.There was no significant difference in average body weight,gastric emptying rate and small intestinal propulsion rate between the groups on the1 st and 7th day after operation(P > 0.05).On the 14 th day after operation,the average body weight,gastric emptying rate and small intestinal propulsion rate of the experimental groups were lower than those of the control group,with statistical significance(P < 0.05).At 1 and 3 months after operation,the average body weight,gastric emptying rate and small intestinal propulsion rate of the long jejunal branch group were lower than those of the short jejunal branch group and the control group(P > 0.05),but there was no significant difference between the short jejunal branch group and the control group(P > 0.05).Six months after operation,there was no significant difference in average body weight,gastric emptying rate and intestinal propulsion rate among the three groups(P > 0.05).The number of ICC cells,the expression of mscf protein and the m RNA expression of c-kit,mscf and Ano1 increased gradually from the first day to 6 months after operation in each group.There was no significant difference in the number of ICC cells,the expression of mscf protein,the m RNA expression of c-kit,mscf and Ano1 in each group on the 1st day after operation(P > 0.05);on the 7th day after operation,the number of ICC cells,the expression of mscf protein,the m RNA expression of c-kit,mscf and Ano1 in each experimental group were lower than those in the control group,and the difference was statistically significant(P < 0.05)There was no significant difference in the number of ICC cells,the expression of mscf protein,the m RNA expression of c-kit,mscf and Ano1 between the experimental groups and the control group at 14 days and 1 month after operation,and there was significant difference in the number of ICC cells,the expression of mscf protein,the m RNA expression of c-kit,mscf and Ano1 among the three groups(P < 0.05)Different).At 3months after operation,the number of ICC cells,the expression of mscf protein,the m RNA expression of c-kit,mscf and Ano1 in the control group were higher than those in the short jejunal branch and long jejunal branch groups(P < 0.05),but there was no significant difference in the number of ICC cells,the expression of mscf protein,the m RNA expression of c-kit,mscf and Ano1 between the short jejunal branch and long jejunal branch groups(P > 0.05).Six months after operation,there was no significant difference in the number of ICC cells,the expression of mscf protein,the m RNA expression of c-kit,mscf and Ano1 among the three groups(P > 0.05).Conclusion: 1.Personalized Roux-en-Y choledochojejunostomy with short Roux branch in children can achieve the same clinical effect as long Roux branch.2.After choledochojejunostomy in young rabbits,the gastrointestinal motility could be restored in 6 months.3.Within 6 months after choledochojejunostomy,the number of ICC cells increased,and the expression of mscf,c-kit,Ano1 gene and mscf protein increased in gastrointestinal tissues.4.The regulation of SCF / c-kit pathway and Ano1 on the number and function of ICC may play an important role in the recovery of gastrointestinal motility.
