Propofol Regulates The Activation Of Schwann Cells By Regulating PTX3

Abstract/Summary:
Objective In this study,the effects of propofol on Schwann cell activation(proliferation and motor migration)by regulating PTX3 were observed in vitro,so as to explore the possible mechanism of propofol promoting regeneration and repair of peripheral nerve injury.Methods The Schwann cells were resuscitated and cultured,and the Schwann cells were silenced by siRNA interference technique.Then,the Schwann cells were divided into blank control group,fat emulsion group,propofol group,propofol +PTX3 silenced group(propofol + Si-PTX3 group),and propofol +PTX3 silenced negative control group(propofol + Si-NC group).(1)CCK-8 method was used to detect the proliferation and activation ability of Schwann cells: cells in each group were seeded on a 96-well plate,and the absorbance value at 450 nm of each well was detected by a microplate analyzer at 48 h.(2)The invasion ability of Schwann cells was detected by Transwell chamber method: cells in each group and DMEM culture medium of 20% serum were placed in the upper and lower layers of Transwell chamber,respectively,and the number of invaded cells was observed and counted for 48 h.(3)The expression of PTX3 in Schwann cells was observed by immunofluorescence method.(4)The expression of PTX3 protein was detected by Western Blot.Results 1.Compared with blank control group,there was no significant difference in fat emulsion group(P > 0.05).Propofol group,propofol + Si-PTX3 group,and propofol +Si-NC group increased Schwann cell proliferation(P < 0.05).Compared with the propofol group,the proliferation of Schwann cells in the propofol + Si-PTX3 group was decreased(P < 0.05),but there was no significant difference in the propofol + Si-NC group(P > 0.05).Compared with propofol +si-NC group,Schwann cell proliferation in propofol + si-ptx3 group was decreased(P < 0.05).2.Compared with blank control group,there was no statistical significance in fat emulsion group(P > 0.05).The number of Schwann cells invasion and proliferation increased in propofol group,propofol + Si-PTX3 group and propofol + Si-NC group(P < 0.05).Compared with the propofol group,the invasion number and proliferation of Schwann cells were decreased in the propofol +si-PTX3 group(P < 0.05),but there was no significant difference in the propofol +si-NC group(P > 0.05).Compared with propofol +si-NC group,the invasion number and proliferation of Schwann cells in propofol + si-ptx3 group were decreased(P < 0.05).3.Compared with blank control group,there was no statistical significance in fat emulsion group(P > 0.05).The Schwann cells were activated significantly in propofol group,propofol + Si-PTX3 group and propofol + Si-NC group,and the number of cells was increased significantly(P < 0.05).Compared with the propofol group,the Schwann cell activation was not obvious and the number of cells decreased in the propofol +si-PTX3 group(P < 0.05),but there was no statistical significance in the propofol +si-NC group(P > 0.05).Compared with propofol +si-NC group,the activation of Schwann cells in propofol + si-ptx3 group was not obvious,and the number of Schwann cells decreased(P < 0.05).4.Compared with blank control group,there was no statistical significance in fat emulsion group(P > 0.05).PTX3 expression was upregulated in propofol group,propofol +si-PTX3 group and propofol +si-NC group(P < 0.05).Compared with the propofol group,the expression of PTX3 in Schwann cells was down-regulated in the propofol +si-PTX3 group(P < 0.05),but there was no significant difference in the propofol +si-NC group(P > 0.05).Compared with propofol +si-NC group,the expression of PTX3 in Schwann cells in propofol +si-PTX3 group was down-regulated(P < 0.05).Conclusion Propofol may promote the proliferation and activation of Schwann cells by regulating PTX3.
