| Objective: How to improve the local micro-environment,inhibit the formation of the glial scar,and promote repair and regeneration of damaged neurons and axons,are the difficulties of treating SCI.The research demonstrated that Ch ABC can enhance the regeneration and plasticity of axons by degrading CSPGs.Meanwhile,BMSCs-exo has a protective effect on SCI recovery by inhibiting apoptosis,regulating immune response,and regulating regeneration of damaged neurons.On this basis,the author combined BMSCs-exo with Ch ABC to explore its synergistic repair effect and mechanism,and provide experimental evidence for exosomes to treat spinal cord injury.Methods : Using the exosomes-extracted kit to isolate BMSCs-exo,observing its morphology under a transmission electron microscope(TEM),and identifying its expressions of CD63 and Alix through western blot.50 SD rats were randomly divided into 5 groups(A: sham group B: model group C: Ch ABC group D: BMSCs-exo group E: Ch ABC+BMSCs-exo group),10 rats in each group.SCI rat models were established by referencing the modified Allen’s method to hit the T10 segment of the spinal cord.The hind limb motor function(BBB)was performed on the 1,3,7,14,and 28 days in each group.After 14 days,each group taken 6 rats to make paraffin slices after perfusion,observed the SCI region by HE staining,and compared the expression of a glial fibrillary acidic protein(GFAP)by immunofluorescence and growth-associated protein-43(GAP-43)by immunohistochemistry.Results:1.Observed under TEM,the appearance of BMSCs-exo is irregular,which is a circular or elliptical shape or “saucer-like” and its size is uneven.The positive expression of its marker proteins CD63 and Alix can be observed in western blot.2.Compared with groups B,C,and D,the E group had a higher BBB score,a smaller damaged area,a lower expression of GFAP,and an up-regulated expression of GAP-43(all P<0.05).Conclusion:Our results demonstrate that BMSCs-exo combined with Ch ABC can inhibit the expression of GFAP in the injured area,up-regulate the expression of GAP-43,reduce the formation of the glial scar,which improve local micro-environment and promote the repair and regeneration of neurons and axons after SCI in rats. |
