| Object:Acute gouty arthritis(AGA)is one of the most common arthroinflammatory metabolic diseases,and its incidence showed a significant upward trend worldwide.However,the pathogenesis of AGA remains unclear,so new metabolic markers are needed for the early prediction and diagnosis.The aim of this study was to reveal serum metabolites changes by UPLC-MS metabonomics from healthy individuals to asymptomatic hyperuricemia(HUA)and then to AGA.Finally,we need to identify potential pathophysiological mechanis ms and biomarkers for gout susceptibility.Method:Firstly,we collected Serum samples from 150 subjects,including 50 healthy volunteers,50 patients with HUA,and 50 patients with gout.Secondly,UPLC-MS metabonomics detection technology was used to obtain metabolic information,and then pair grouping statistics was conducted.Principal component analysis(PCA),partial least squares discriminant analysis(PLS-DA)and T test were used to obtain statistically significa nt difference metabolites.Thirdly,the selected metabolites were applied as early diagnostic markers for HUA and AGA patients,and the metabolic pathways involved were identified by retrieval in the KEGG database.Finally,combined with relevant literature,the metabolic pathway map was drawn.Result:Seven serum metabolic markers that could trigger the onset of HUA and AGA were detected by UPLC-MS metabonomics.They were Lyso PC(16:0),Lyso PC(18:0),Lyso PC(22:6(4Z,7Z,10 Z,13Z,16 Z,19Z)),PC(17:0/0:0),pantothenic acid,phenylpyruvate,and SM(D18:1/22:0),which were involved in two metabolic pathways: phospholipid metabolis m,apoptosis metabolism and choline metabolism.In addition,the two distinct metabolites of Lcarnitine and 10-deoxybacolactone were only induced in the serum of AGA patients.Pathway analysis showed that the enriched metabolic pathway was energy metabolism.The other two distinct metabolites of Methylhistidine and quinolinic acid were only selected in the serum of HUA patients,and the metabolic pathways involved are amino acid metabolism and purine metabolism.The possibility of these metabolic changes brings new hope for the early prediction and diagnosis of AGA.Conclusion:In conclusion,UPLC-MS analysis was performed on 150 serum samples from HUA group,AGA patients and healthy control group,and it was finally inferred that metabolic pathways such as apoptosis metabolism,choline metabolism,lipid metabolism,amino acid metabolism and energy metabolism were disordered in HUA and AGA patients.Differe nt metabolites and metabolic pathways suggest that disease progress leads to changes in biomarkers,which may provide screening targets for early diagnosis and treatment. |
