Objectvie: To explore whether there are differences in the expression levels of mi R-545-3p and LRP5 in serum and bone marrow between patients with senile osteoporosis and normal controls.Methods: Nineteen patients with hip fractures(including femoral neck fracture and intertrochanteric fracture)in the first affiliated Hospital of Anhui University of Chinese Medicine from September 2020 to September 2021 were selected,including femoral neck fracture treated with hip arthroplasty and femoral intertrochanteric fracture treated with intramedullary fixation.Patients with diabetes,severe renal insufficiency,hyperthyroidism,hypothyroidism,blood diseases,malignant tumors and other diseases affecting bone metabolism in the past 5 years were excluded.Twelve patients whose preoperative bone mineral density(BMD)was less than-2.5 and diagnosed as osteoporosis were taken as the experimental group.Seven patients with no osteoporosis and T value ≧-1.0 were included in the control group.The experimental specimens are the patient’s serum and bone marrow fluid,the serum comes from the peripheral blood exudated from the visual field of the patient during the operation,and the bone marrow fluid comes from the reaming process of the patient during the operation.Firstly,RNA was extracted from serum and bone marrow fluid samples,and then c DNA was obtained by reverse transcription RNA.Then the expression levels of mi R-545-3p and LRP5 in serum and bone marrow tissue of the two groups were detected by real-time fluorescence quantitative method(RT-q PCR).Finally,statistical analysis and drawing were carried out by SPSS22.0 software and Graph Pad Prism8 software to analyze the data difference between the two groups.Results: The expression level of LPR5 in the bone marrow of the experimental group was lower than that of the control group,and the difference was statistically significant(P<0.05).Conclusion: Compared with the normal population,the expression level of LRP5 in the bone marrow of patients with osteoporosis is lower,and the decreased expression level of LRP5 may indicate the occurrence of osteoporosis in humans.
